摘    要:对大球盖菇(Stropharia rugoso-annulata)原生质体制备的条件进行了研究,并确定了最佳条件组合:选用15 g/L的纤维素酶+15 g/L的蜗牛酶的混合酶来酶解菌丝,0.6 mol/L的甘露醇作为渗透压稳定剂,菌龄为3d,酶解温度30℃,酶解时间2.5h,在此条件下制得的原生质体产量最高达1.70×...

Abstract:Conditions of preparating protoplast of Stropharia rugoso-annulata were studied.The result shows that the highest production rate of protoplast(1.70×107 mL-1) can be obtained on these conditions:3-day-old mecia,15 g/L cellulase and 15 g/L snailase admixture,dissolution in 0.6 mol/L mannitol,resolved at 30 ℃ for 2.5 h.Ultraviolet radiation inducing S.rugoso-annulata protplast was researched too.The conclusion is that 35 s is the irradiation time,deadly ratio is 73.33%.
