Abstract:In order to confirm that the chicken embryonic lens in vivo is a useful experimental system, we carried out a comparative study on morphology of chicken embryonic lenses in rive and in vitro with the same experimental treatment. Monoclonal antibody NC6 and tumor promoter TPA (12-tetradecanoyl phorbol-13-acetate), which have been proved to make lenses bigger, were used as the experimental agents, and chicken embryos at stage 20 were used as the experimental materials. For the experiment of lenses in vivo, NC6 or TPA was injected into right eyes while uninjected left eyes served as the control. For that in vitro, lenses of the experimental group were cultured in the media with NC6 or TPA. and those of the control group in the media without NC6 or TPA. The size of lenses was measured and their structure was observed after the treatment for 24 hours. The results of lenses in vivo indicated that the size of the lenses from the experimental group is obviously larger than that from the control group (P<0.01), but no visible change in their structure could be found in the experimental group. The experiment of lenses in vitro shows the same results. It is well known that the development of lenses in vitro is not subjected to effects of any mechanism within embryos. The fact that the result of lenses in vivo is the same as that in vitro confirms that chicken embryonic lens in vivo is a useful experimental system. This system seems to be very suitable for a detailed analysis of some important questions, such as the growth control, effects of chemicals on development etc. The project was supported by Grants from NSF of China and SM of Jiangsu Province.
