多重响应性羧甲基壳聚糖智能水凝胶的制备与其响应性研究

摘    要:智能凝胶的交联及其响应性对药物(尤其是多肽或蛋白类)的活性及靶向传递有着重要影响.该文采用巯基化的羧甲基壳聚糖实现温和条件制备多重响应性的智能水凝胶;研究凝胶的溶胀行为、盲结肠酶的降解行为和谷胱甘肽的还原响应性行为.凝胶显示出pH敏感、酶敏感和还原敏感性. 巯基化羧甲基壳聚糖的巯基含量及凝胶的交联程度不仅影响凝胶的溶胀行为,而且影响凝胶的酶降解性;凝胶的还原敏感性依赖于凝胶的交联程度.凝胶显示出可用作结肠靶向给药系统载体传递多肽或蛋白类药物的潜力.

Abstract:The cross-linking of intelligent hydrogels and their responsiveness have an important effect on the activity of drugs (especially polypeptide or protein drugs) and controlled release. In this paper, mercapto-carboxymethyl chitosan was used to prepare the multi-responsive intelligent hydrogels. The swelling behavior of the hydrogels, the degradation behavior by the colonic enzyme and the reduction behavior by glutathione were studied. The hydrogels showed pH sensitivity, enzyme sensitivity and reduced susceptibility. The thiol content of thiolated carboxymethyl chitosan and the degree of cross-linking of hydogel not only affect the swelling behavior of the hydrogels, but also affect their enzymatic degradation. The reducing sensitivity of the hydrogels depends on their degree of disulfide cross-linking. The hydrogels show the potential for the delivery of polypeptides or protein drugs to colon system.
