| 摘 要： | 为了获得高质量的艾美耳属（Eimeria）球虫卵囊基因组DNA，采用了4种不同方法提取3种艾美耳属球虫卵囊的基因组DNA．对比结果表明，采用机械法破裂球虫卵囊壁，用裂解液裂解孢子囊和子孢子，释放出DNA，在保持碱性的环境下（pH=8．0），以苯酚-氯仿异戊醇抽提，乙醇沉淀，可获得高质量的DNA分子，经PCR检验为艾美耳属球虫卵囊基因组DNA．

| Abstract: | The goal of the experiment is to find out which method can produce high-quality Eimeria genomic DNA.Four different methods were used in the study.The results showed that highly pure DNA molecules was obtained by the mechanical method.Mechanical breakdown was used to break the oocyst-wall and lysis buffer was used to dissolve sporocysts and sporozoites to release DNA.DNA was extracted with phenol-chloroform-isoamyl alcohol while maintaining the basic environment(pH=8.0) and was precipitated by ethanol.The su... |
