| 摘 要： | :用逆相法、脂膜法制得了禽流感病毒血凝素 (HA)基因重组质粒pSVH7DNA(核酸疫苗 )脂质体 ;用荧光法测定了质粒DNA脂质体的包封率 ,两种方法所得脂质体的包封率分别为 15 .5 8± 1.5 5 % ,6.0 8±0 .3 5 % ,当在磷脂成份中加入阳离子十八胺时 ,脂质体的包封率分别为 70 .4 1± 6.4 9% ,4 6.5 6± 1.92 % 磷脂浓度和离子强度均能影响脂质体的包封率 ,前者能增加包封率 ,而后者使其减少 电镜结果表明 ,两种方法所得的脂质体为单室或多室球形脂质体混合物 ,其粒径大小分别为 163nm和 2 3 5nm

| Abstract: | AIVHAgeneexpressingplamidpSVH7DNAliposomeswerepreparedbyreversephaseevaporation(REV)andthinfilm(TF)method .TheDNAencapsulationyieldwas15 .5 8± 1.5 5 % and 6.0 8± 0 .35 %respectivelyasmeasuredbyethidiumbromidefluorescencespectramethod . Additionofacationicstearylamine(SA)inliposomecompositionincreasedtheencapsulationyield upto 70 .41± 6.94%and 46.5 6± 1.92 %respectively .Thelipidconcentrationandionic strengthofthesolutionaffectedtheencapsulationefficiencygreatly ,theformerincreasedthe yieldwhilethelaterdecreasedit.Electronmicroscopeobservationshowedthattheliposomesus pensionwasmixtureofsmallunilamellarvesiclesandlargermultilamellarvesicleswhichconsisted ofseveralconcentricbilayers.Theaveragediameteroftheliposomewas 163nmand 2 35nmre spectively . |
