黑稻蛋白快速测定法初探

摘    要:本文用凯氏定氮法为标准衡量考马斯亮蓝染色法和碱式碳酸铜法的测定结果,其中考马斯亮蓝染色法测定黑稻蛋白含量时,不受色素影响,结果平均偏差-0.027mg/g,而碱式碳酸铜法,则受色素影响稍大,结果平均偏差0.259mg/g。同时,专门对前法中染液不同H3PO4终浓度及不同蛋白提取条件对结果的影响作比较,确定H3PO4在染液中终浓度为12%(W/V)俚,可获一通过零点的标准曲线,而用85%-95%乙醇对材料蛋白提取10分钟,是获得较准确数据的提取方法。

Abstract:Pigments in the grain of black rise ( Oryza sativa) interfere with the accurate measurement of protein content by means of alkaline copper carbonate. The method of Coomassie brilliant blue and that of alkaline copper carbonte are Compared. Judged by Micro-Kjaldahl's method,results indicate that the former is not influenced by the pigments, its deviation being-0.027mg/g; while the latter is greatly influenced, its deviation being +0. 259mg/a The conclusion is in favour of Coomassie brilliant blue. The standard procedure of determination, protein in grain extracted with 85-95% alcohol for 10 minutes;terminal H3PO4 concentration in staining solution is 12%(W/V). This standard procedure can obtain a working curve through zero point.
