大蒜素纳滤提取及对粪肠球菌的体外抑菌研究

摘    要:以白皮大蒜为原料,研究大蒜素的纳滤提取条件和大蒜素对粪肠球菌Enterococcus faecalis的体外抑菌特性.研究结果表明:0.45 μm膜过滤后的大蒜液在压力0.6 MPa、温度36℃操作条件下经孔径300 Da的纳滤膜纳滤后,大蒜原料中大蒜素的有效提取率为81.9%,纳滤液中大蒜素含量为2.07 mg/mL.大蒜素对E. faecalis的最小抑菌浓度和最小杀菌浓度在好氧和厌氧培养条件下无明显差异,分别为15 mg/mL和23 mg/mL.20 mg/mL的大蒜素对E. faecalis

Abstract:The optimum extractiing conditions of allicin from Allium sativum L. by nanofiltration membrane were investigated, as was also its inhibition against Enterococcus faecalis in vitro. The results showed that the extraction rate of allicin was 81.9% under 300 Da nanofiltration, pressure 0.6 MPa, temperature 36 ℃ after allium bulbs were crushed and filtered by 0.45 μm microfiltration. And allicin in the solution transported by nanofiltration was 2.07 mg/mL. The extract's minimum inhibitory concentrations (MIC) against E. faecalis in vitro had no differnce when 37 ℃ aerobic and anaerobic environment respectively for growth by the broth microdilution method, as aslo to its minimum bactericidal concentrations (MBC). Its MIC and MBC were 15 mg/mL and 23 mg/mL, respectively. Agar diffusion tests showed that the nanofiltration extraction liquids contained 20 mg/mL allicin produced zone diameters of inhibition 12 mm~33 mm and the distribution of all zones was normal (Sig>0.05) by Kolmogorov-Smirnov test used SPSS 15.0. Furthermore, the median of 20 mg/mL allicin in boxplot was 24 mm more than it of an activity equivalent to 15 mg/mL sodium hypochlorite with exploratory data analysis by SPSS 15.0.
