| 摘 要： | Canstatin是最近发现的能抑制新生血管生成和肿瘤生长的又一血管生成抑制因子。从胎盘组织中提取总RNA，根据已知canstatin基因序列，设计特异引物，应用RT－PCR方法扩增出该基因片段。DNA序列测定表明，与文献报道的该基因比较，核苷酸序列完全一致。将扩增得到的该基因克隆于原核表达载体pPROEX-HTb中，在大肠杆菌E;coli BL21中经IPTG诱导获得了表达，表达量约占菌体总蛋白量的20％以上。

| Abstract: | Canstatin is one of recently discovered angiogenesis inhibitor which can inhibit angiogenesis and tumor growth.Total RNA was extracted from human placenta tissue and canstatin gene was amplified by RT-PCR.The result from nucleotide sequencing indicated that it is the same as canstatin sequence reported.The cansatin gene was then cloned into prokaryotic expression vector pPROEXHTb and overexpressed in E.coli BL21. |
