摘    要:用响应面方法对红豆杉细胞培养生产紫杉醇的几种前体及诱导子进行了优化,首先,用部分因素实验对苯丙氨酸,脱落酸,硝酸银,苯甲酸钠,醋酸钠,壳聚糖,甘氨酸对紫杉醇含量的影响进行评价,并找出其中主要影响因子壳聚糖,硝酸银和脱落酸;其次,用最速上升法逼近最大响应区,最后用中心组合设计和响应面分析确定主要影响因子的最适浓度,经优化后的紫杉醇体积质量达到60.5mg/L,较优化前提高1倍,利用以上结果进行5L反应器放大培养,紫杉醇体积质量达54.6mg/L。

Abstract:The concentration of several precursors and elicitors for taxol production of Taxus Chinensis is optimized by means of response surface method. The effect of phenylalanine, abscisic acid, sliver nitrate, sodium benzoate, sodium acetate, chitosan and glycine on taxol production was evaluated by using a fractional factorial design. The major factors were proved to be chitosan, sliver nitrate and abscisic acid. After approaching the optimal region along the path of steepest ascent, a central composite design and corresponding response surface analysis were used to obtain the optimal concentrations of the major factors. Taxol production could reach 60.5?mg/L after optimization, which was once higher than taxol production without optimization. When the process was scaled up in a 5?L bioreactor, taxol production of 54.6?mg/L was obtained.
