生姜蛋白酶的分离纯化及其糖肽键型的初步研究

摘    要:本文报导以生姜为材料,冷丙酮脱脂脱色后,用0.1mol/LPH6的磷酸缓冲液提取、硫酸铝盐析,经DEAE-纤维素DE-52和SephadexG-75纯化,得生姜蛋白酶两个组份ZE1和ZE2。它们经凝胶等电聚焦电泳,均为单一的蛋白带,PI值别为7.0和8.0。苯酚-硫酸法鉴定,其均为糖蛋白。β-消去反应表明:ZE1和ZE2中的糖肽键型均为非0-型糖肽键。

Abstract:Zingiber offcinai Roscoe defatted with cold acetone was extracted with 0. 1 mol/L phosphatebuffer (PH6. 0). The obtained rough Zingiboin was pun floatal by ammonium sulfate salting-out,DEAE-celluloce DE-52 column chromatopaphy and SephadexG-75 chromatography, and two proteinases, ZE1 and ZE2,were gumed. Isoelectrofocusiflg electrophoreds proved both of them were homogeneous,and revealed the PI Values were 7. 0 and 8. 0 respectively. By means of phenolsulfuric acid methed,both ZE1 and ZE2 were identified to glycoproteins. The glycsidic linkages of ZE1 and ZE2 were shown to be non--o-type ones.
