摘    要:采用高聚物反相填料装填的半制备柱(PST, 30μm, 300mm×10mm ID)对莱菔硫烷进行分离纯化,优化了色谱分离参数,确定最佳流动相为30%乙腈-水溶液,流速为2mL/min等度洗脱,检测波长254nm,进样量不超过20mg时,收率在61%以上,纯度可达90%。采用中心切割的方式收集,获得的产品纯度可达90%以上。

Abstract:Sulforaphane has been separated and purified by polystyrene-based reversed phase chromatography(PST,30 μm,300 mm×10 mm ID).The preparative separation conditions were optimized and the optimum parameters were found to be 30% acetonitrile in water as the mobile phase,and a flow-rate of 2 mL/min in the isocratic elution mode.The highest purity of sulforaphane was 90% with a loading of 20 mg.The yield was more than 61%.With the use of overloading conditions and the heart-cutting method,the purity of sulforaphane exceeded 90%.
