蚕豆外源凝集素的研究

摘    要:用葡聚糖凝胶G—75亲和层析柱和猪甲状腺球蛋白—ABSE交联琼脂亲和层析柱纯化所得的蚕豆外源凝集素,其聚丙烯酰胺凝胶电泳图谱显示:前一方法提取的蚕豆外源凝集紊呈一条深色带,一条浅色带,后一方法提取的蚕豆外源凝集素呈一条显色带。经SDS—聚丙烯酰胺凝胶电泳测定其亚基分子量为23800。血凝反应表明蚕豆外源凝集素能显著地凝集兔的红细胞,对人(A型和O型)和猪的红细胞也有不同程度的凝集效应,并对人的外周血淋巴细胞具有促有丝分裂作用。用猪甲状腺球蛋白—ABSE交联琼酯作为亲和吸附剂与国外沿用的Sephadex G—75法相比较,具有成本低、操作步骤较简便的优点,此外,它所提取的蚕豆外源凝集素的生物活力也较鬲。


STUDIES ON LECTIN FROM VICIA FABA
Abstract:We have purified fava lectin by Sephadex G—75 and procine thyroglubulin—ABSE p—Aminobenyene sulphonyl ethyl—(ECD)]—cross—Linked agar as affinity absorbent respectively. The fava Lectin by the former Show two protein bands, a major band, a minor band, and the fava Lectin by the latter has a protein band on polyacrylamide gel electrophoresis. Its subunit molecule weight were estimated to be 23,800 by SDS—polyacrylamide gel electrophoresis. The fava Lectin can agglutinined rabbit erythrocytes remarkably, It also agglutinated human (A,B) and procine erythrocytes in varying degrees. At the same time, It is also rnitogenic for human Lymphocyte. In Contrast to sephadex G—75 as affinity absorbent, procine thyroglubulin—ABSE—cross—Linked agar as affinity absorbent has not only lower product cost but also simple operate method. Moreover, The fava Lectin purified by the method has higher biological activity,
