Abstract:The orthogonal test with factors of 0.05-0.4 mg/L 6-BA and 0.05-0.2 mg/L NAA/IAA/IBA was taken for the microapical culture of daphen odora var.marginata.The results showed that the bud forming rate was the highest in the combination of 0.05-0.2 mg/L 6-BA and 0.05-0.1mg/L IBA.The bud forming rate was related to the size of apex and the number of leaf primordium.The bud forming rate was low(34%) as the size of apex was small with few number of leaf primordium,but the detoxification rate was high(90%).By the examinations of biology,serology and electron microscope,it was found that 5 kinds of virus of daphen odora var.marginata were debuged from the plantlets.In the test of proliferation with different concentrations of 6-BA,it was found that the combination of 0.5 mg/L 6-BA and 0.1 mg/L NAA was the best,in which the buds were strong and the propagation coefficient was high(3.17).In the test of proliferation with different parts of the shoot,it was found that the apex was better than the middle and end parts of the shoot,which was with the highest propagation velocity,pecentage of effective advetitious bud and qulity of bud.
