To find out whether physiological concentrations of cholecystokinin (CCK), a gastrointestinal hormone in mammals, are also active in chickens, the pancreatic amylase secretory response to CCK-8 was investigated in vitro. Rat pancreatic acini responded to the physiological concentration of CCK-8, but in chickens amylase release was induced at a concentration of CCK-8 1000 times higher than that observed in rats. In another experiment, biliary flow was tested with several doses of CCK-8. The bile flow was stimulated in a dose-dependent fashion, but a significant enhancement was not obtained at a concentration of 0.5 g CCK-8/kg body weight, which was considerably higher than physiological ones. It is concluded that endogeneous CCK does not have an important role in the digestive system in the chicken.  相似文献

本文建立了用荧光指示利Fura-2测定胰腺腺泡细胞胞浆游离Ca2+浓度([Ca2+]i)的实验技术,包括:胰腺腺泡的分离制备;Fura—2/AM的负载;腺泡悬液的荧光光谱测量等.测定了胰腺腺泡细胞[Ca2+]i。的静息值和卡巴可促腺泡细胞[Ca2+]i升高的剂量—效应关系及动力学过程,并研究了柴胡总皂甙对胰腺腺泡细胞[Ca2+i]的影响.发现0.01mg/ml浓度的柴胡总皂甙使胰腺腺泡细胞[Ca2+]。明显升高,0.1mg/ml浓度时可使[Ca2+]。升高到静息值的3倍,提示柴胡总皂甙的促酶分泌作用是通过胞内[Ca2+]i升高介导的.  相似文献

Pteroyl polyglutamate hydrolase (folyl conjugase), which hydrolyses the dietary polyglutamyl folates into simple forms prior to absorption, has been shown to be induced in rat pancreas in response to dietary polyglutamyl folates but not by ingestion of synthetic unconjugated folates. Folate absorption, as measured by the rise in serum folate levels after ingestion, of dietary conjugated folates is impaired in pancreatectomised animals whereas absorption of synthetic simple folate is not. A severe build-up of folyl conjugase is observed in the lumen of control but not in pancreatectomised animals after dietary folate ingestion. These results taken together would suggest that dietary folates are hydrolysed to monoglutamyl forms suitable for absorption in the lumen; the hydrolysis is catalysed by a luminal folyl conjugase of pancreatic origin induced by dietary conjugated folates.  相似文献

枸杞多糖对Ⅱ型糖尿病小鼠胰岛细胞形态与功能的影响   总被引:3,自引:0,他引:3  
采用高糖高热量饮食并结合小剂量腹腔注射STZ诱导Ⅱ型糖尿病(NIDDM)小鼠模型,枸杞多糖灌胃4周,用免疫组化技术对胰岛A、B细胞进行形态学分析及观察血糖、血清胰岛素的变化。结果显示,枸杞多糖大,小剂量组和模型组比较空腹血糖下降,胰岛素水平升高。图像分析大剂量组和模型组比较,B细胞数密度增加、A细胞数密度减小、B细胞切面积、核面积、核质比升高。结论:枸杞多糖可以改善Ⅱ型糖尿病小鼠胰岛细胞形态和功能并促进胰岛素的分泌,有良好的降血糖作用。  相似文献

Summary Mechanisms by which pancreatic secretions influence disaccharidase activities in the distal small intestine have been investigated in 1-week-old miniature pigs. Using a combination of biochemical, cytochemical and morphological techniques it has been found that the decrease in lactase specific activity is due solely to a reduction in villus surface area. By contrast, the increased sucrase-isomaltase activities, which occur despite the reduction in villus surface area, are due entirely to increased enzyme expression during enterocyte differentiation.  相似文献

The present study was done to investigate the factors regulating the intestinal phase of exocrine pancreatic secretion in the turkey. The intestine of turkeys equipped with pancreatic fistulas was perfused with peptone solution, fat emulsion and hydrochloric acid (HCl), and pancreatic flow and protein output were measured. Neither peptone solution nor fat emulsion had any effects on pancreatic secretion. HCl enhanced the flow rate of pancreatic juice but not protein output. To clarify the neural mechanism of this phenomenon, the vagal postganglionic blocker atropine was continuously infused and pancreatic secretion in response to intestinal HCl was measured. Atropine completely suppressed both pancreatic flow and protein output. It is suggested that the avian intestinal phase of pancreatic secretion is mainly controlled by cholinergic action though HCl stimulation.  相似文献

We investigated the effect of aging on glucose uptake, glucose-induced O2 consumption, glucose-induced45Ca movements, and calmodulin content to elucidate age-related impairment of glucose-induced insulin release in pancreatic islets of Wistar rats. Intact pancreatic islets from old (24-month-old) rats showed impaired glucose-induced insulin release; glucose uptake and O2 consumption were lower in old than in young (2-month-old) or adult (12-month-old) rats. Moreover,45Ca uptake and calmodulin content were decreased in pancreatic islets from older rats, which explained the impairment in glucose-induced insulin release in aging. No major differences between the 3 age groups in glucose-induced45Ca efflux in pancreatic islets were observed.  相似文献

采用葡萄糖和棕榈酸钠(Glu-Pal)联合诱导大鼠胰岛瘤RINm-5F细胞凋亡,用噻唑蓝比色(MTT)实验检测次血红素六肽(DhHP-6)和Exenatide分别或联合作用对细胞凋亡的影响.结果表明:DhHP-6和Exenatide均显著抑制Glu-Pal诱导β细胞凋亡,但二者无协同作用;DhHP-6和Exenatide均明显促进RINm-5F细胞增殖,且有显著的协同作用.  相似文献

胰激肽原酶片对早期糖尿病肾病疗效观察   总被引:1,自引:0,他引:1  
目的:评价胰激肽原酶片治疗早期糖尿病肾病患者的疗效和安全性.方法:将82例早期糖尿病肾病患者随机分成2组,采用糖尿病饮食控制、体育锻炼及根据病情给予口服降糖药物或/和胰岛素治疗,治疗组(42例)加用胰激肽原酶片治疗4周.观察治疗前后尿微量白蛋白与24h尿蛋白定量和不良反应.结果:治疗组治疗前后尿微量白蛋白与24h尿蛋白定量明显降低,差异有统计学意义(P<0.01);对照组治疗前后比较差异无统计学意义(P>0.05);组间比较差异有统计学意义(P<0.01);2组患者无明显的药物不良反应.结论:胰激肽原酶片治疗早期糖尿病肾病患者疗效好,用药安全,可广泛应用于临床.  相似文献
