Summary Human fibroblasts and A431 human epidermoid carcinoma cells were cultured on gamma-irradiated human amnion collagen as well as on plastic dishes and non-irradiated collagen coated dishes. The morphology, attachment, growth and short-term cytotoxicity of these culture conditions have been determined. Both irradiated and non-irradiated amnion collagen enhanced the attachment and proliferation of fibroblasts as compared to the plastic dishes. No differences in these properties were observed for A431 cells cultured on irradiated collagen when compared with culture on non-irradiated collagen substrates. Cytotoxicity assays showed that irradiated and non-irradiated collagens were not cytotoxic for either fibroblasts or A431 cells. The results demonstrated that amnion collagen irradiated at doses of 0.25–2.0 Mrads is optimal for cell growth.  相似文献

研究了聚富马丙二醇酯(PPF)的合成方法,讨论了合成条件(催化剂种类、用量、反应时间)对合成反应的影响,发现最佳反应条件为富马酸与丙二醇物质的量之比为1:1,对甲苯磺酸与富马酸的物质的量之比为2.33:100,在90℃下反应7h。利用FTIR,TGA,GPC等方法对其进行了表征。研究了这种树脂的自由基交联和光固化性能,发现交联时最高放热温度为36℃。  相似文献

新型可注射温敏水凝胶的制备及其释药性能   总被引:4,自引:0,他引:4  
采用改性壳聚糖作为药物载体制备了一种可注射温敏水凝胶。将单甲氧基醚聚乙二醇2000(mPEG2000)接枝到壳聚糖(CS)上制备壳聚糖接枝产物(mPEG-g-CS);将接枝产物在一定浓度下配制成凝胶溶液,测定其温敏性能;以温敏水凝胶为载体,以布洛芬为模型药物进行体外释放研究。结果表明,mPEG-g-CS水凝胶在体温37℃附近发生溶胶到凝胶的转变,具有良好的温敏性能,在一定的浓度范围内提高mPEG-g-CS浓度可降低相变温度,缩短胶凝时间。体外释药结果表明mPEG-g-CS水凝胶对模型药物具有缓释作用,有望用作可注射温敏型药物控释载体。  相似文献

A new type of injectable premixed bone cement consisting of nano-hydroxyapatite (n-HA) and polyamide 66(PA66) composite is investigated.This cement can be handled as paste and easily shaped.which can set in air,in physiological saline solution and in blood.The setting time,injectability and compressive strength of the cement largely depend on the ration of liquid to powder (L/P),Moreover,the content of n-HA in composite also affects the compressivce strength and injectability of the cement.The premixed composite cement can remain stable in the package for a long period and harden only after delivery to the defects site.The results suggest that injectable premixed cement has a reasonable setting time,reasonable viscosity for injecting,excellent washout resistance and high mechaical strength,which can be developed for root canal filling,sealing and various bone defects augmentation.  相似文献

将N-亚甲基磷酸化壳聚糖(NMPC)引入海藻酸钙水凝胶体系,制备了力学强度和稳定性均优于海藻酸钙和海藻酸/壳聚糖复合水凝胶的可注射型材料.通过正交实验,根据材料的力学性能、浸提液pH值和凝胶化时间的对比分析,确定了各组分质量分数为:海藻酸钠1.50%、碳酸钙0.30%、N-亚甲基磷酸化壳聚糖1.00%、柠檬酸0.30%.细胞培养和肌内植入试验表明,海藻酸/磷酸化壳聚糖(Alg/NMPC)复合水凝胶具有良好的生物相容性.  相似文献

可注射水凝胶作为生物材料被广泛用于生物医学领域. 合成了含有二硫键的葡聚糖衍生物(Dex-SS-PA), 使用辣根过氧化酶化学交联制备出新型的生物还原响应的可注射水凝胶. 系统研究了这些水凝胶的物理化学属性, 包括成胶时间、胶含量及降解性, 同时使用生死染色实验和噻唑蓝比色法实验评价了它们的细胞生物相容性. 研究结果表明, 葡聚糖的分子量越大、根皮酸的取代度越高、Dex-SS-PA 的浓度越大, 获得的水凝胶成胶时间越短、胶含量越高. 在还原环境中, Dex-SS-PA 水凝胶能够被降解. 此外, 这些水凝胶没有明显的细胞毒性. 因此, 这类新的水凝胶预示着较好的生物医学应用前景.  相似文献

Chemically cross-linked hydrogels constitute a novel injectable tissue engineering material. At present, one of the key problems is to find an appropriate initiator. This study evaluated the cytotoxicity in vitro of a water-soluble redox initiating system consisting of ammonium persulfate (APS) and N, N, N‘, N‘-tetramethylethylenediamine (TEMED). Gelation time of PEG diacrylate macromer in phosphate buffer saline solution was first adjusted to guarantee that the examined initiator concentrations are sufficiently high to trigger polymerization of macromers. NIH/3T3 fibroblasts were employed to examine cytotoxicity via MTT measurements and optical microscopic observations. It has been found that the combined APS/TEMED system exhibits negative cooperative effect, for the underlying cytotoxicity is even lower than that of APS or TEMED at certain concentrations.  相似文献

可注射生物活性陶瓷不仅具有良好的生物相容性,而且能够促进成骨分化和血管生成。羟基磷灰石(Ca10(PO4)6(OH)2, HA)是人骨的主要成分。采用微流控技术,成功制备出粒径480 μm的均一硅酸钙(CaSiO3, CS)微球,并以其为前驱体,在磷酸三钠(Na3PO4)水溶液中进行24 h水热转化处理,得到具有纳米线结构的含硅羟基磷灰石微球(Si-nHA)。该纳米线直径约100~200 nm,长度约1~2 μm。Si-nHA微球比表面积为7.78 m2/g,远高于纯HA微球的比表面积(0.08 m2/g),高比表面积使得Si-nHA微球具有更加优异的蛋白负载和缓释性能。同时,Si-nHA微球释放的硅(Si)离子能显著促进大鼠骨髓间充质干细胞(rBMSCs)的增殖,以及相关成骨(OPN,OCN)和成血管基因(ANG-1)的表达。研究结果表明,Si-nHA微球在微创骨修复领域有良好的应用前景。  相似文献
