利用荧光标记的PCR法,对唐氏综合症的检验进行研究.采用法医学检验常用试剂盒AmpFe STR^TM Profiler Plus^TM对唐氏综合症病例进行PCR扩增,扩增产物用ABIPRISMT”310型DNA全自动测序仪分型判断.结果表明,该检验方法具有简便、快速、直接、准确的特点,检验效果非常好,而且可用于产前诊断.该方法用于检验唐氏综合症是一种理想的方法. 相似文献

Summary The binding of amiodarone to human plasma protein and to bovine serum, albumin was studied by three different methods, ultracentrifugation, equilibrium dialysis and fluorescence spectroscopy. The fraction of amiodarone bound to plasma protein amounted to 96.3%. The changes in the binding properties of 1-anilino-naphthalene-8-sulfonate for bovine serum albumin using warfarin and amiodarone as independent inhibitors were analyzed in terms of binding site specificity. The findings indicated that amiodarone and warfarin have two different binding sites on bovine serum albumin, so a noncompetitive inhibition mechanism was indicated. On the basis of our data we cannot exclude other mechanisms of interaction besides direct displacement of one drug by another; nevertheless, metabolite interference between amiodarone and coagulation cofactors may better explain the enhancement of warfarin's pharmacological action in association with amiodarone.This work was partially funded by the CNR (National Research Council, Rome, Italy), Program on Clinical Pharmacology and Rare Diseases. Summary Visual determination of MSH-induced pigment migration in melanophores of small pieces ofAnolis carolinensis skin is standardized by first measuring photoelectrometrically the change in reflection/transmission of the whole dorsal skin in response to different hormone concentrations. This method allows the rapid and precise recording of time-response curves after photoaffinity labeling of MSH receptors or of dose-response curves of large series of synthetic compounds.Acknowledgments. A method for real time observation of photo-cleavage of stratched λDNA at single molecular level by a fluorescent microscope coupled with CCD is developed.DNA molecules stained with YOYO-1 are stretched by the mo-lecular combing technique and fixed on a modified slide.Then the Process of Photocleavage and relaxation of DNA under radiation of blue light is observed.We speculate that the conformation change of stretched DNA and the effect of water are likely to facilitate the effect of YOYO photocleav-age DNA molecules.The photocleavage effect of YOYO for stretched DNA may be useful to study DNA elasticity,cancer research as well as the interaction between DNA and dyes. 相似文献

研究一个烟草花叶病毒(TMV)cDNA3′末端被特异切割的发夹核酶对感染原生质体的作用。选用一个外源基因绿色水母荧光蛋白(GFPuv)作为报道基因,取代了大部分的CP编码区域,以研究在烟草原生质体中发夹核酶对TMV载体的复制,通过GFP的表达来确定表达量。顺式发夹核酶对TMV载体感染效率的影响通过体外转录来观察。TMV GFPRIB体外转录感染原生质体的效率比与其不含该核酶的对应物的感染效率高3~5倍。Northern杂交结果表明,包括核酶在内的3’末端非病毒序列在体外转录反应开始后就被顺式发夹核酶切除了。当直接用载体DNA pSTMVGFPRIB感染原生质体时,感染原生质体的数量比与之相对应的pSTMVGFP NOS感染原生质体数多50%~80%。 相似文献
