Toll-like receptors (TLRs) act as sensors of microbial components and elicit innate immune responses. All TLR signaling pathways
activate the nuclear factor-kappaB (NF-κB), which controls the expression of inflammatory cytokine genes. Transforming growth
factor-β-activated kinase 1 (TAK1) is a serine/threonine protein kinase that is critically involved in the activation of NF-κB
by tumor necrosis factor (TNFα), interleukin-1β (IL-1β) and TLR ligands. In this study, we identified a novel protein, WD40
domain repeat protein 34 (WDR34) as a TAK1-interacting protein in yeast two-hybrid screens. WDR34 interacted with TAK1, TAK1-binding
protein 2 (TAB2), TAK1-binding protein 3 (TAB3) and tumor necrosis factor receptor-associated factor 6 (TRAF6) in overexpression
and under physiological conditions. Overexpression of WDR34 inhibited IL-1β-, polyI:C- and lipopolysaccharide (LPS)-induced
but not TNFα-induced NF-κB activation, whereas knockdown of WDR34 by a RNA-interference construct potentiated NF-κB activation
by these ligands. Our findings suggest that WDR34 is a TAK1-associated inhibitor of the IL-1R/TLR3/TLR4-induced NF-κB activation
pathway.

A mutant strain of E.coli showing strong green fluorescence is obtained during the study of cloning and expression of blue fluorescent protein gene bfp. A recombinant plasmid pHN122 has been isolated. It is demonstrated by enzyme digestion, subcloning and sequence analysis that it contains both of bfp and gfpK79R mutant genes. The results of spectrometric analysis show that GFPK79R expressed by pHN122 is the same as the wild type GFP, but its fluorescent intensity is increased 1.25~2.44-fold. 相似文献

人WDR79是一种重要的支架蛋白,在端粒酶组装、卡哈尔体(Cahar body)形成和DNA损伤修复等过程中发挥着重要作用。采用PCR扩增技术获得人WDR79基因启动子上游DNA序列,构建人WDR79基因启动子荧光素酶报告基因载体pGL3-WDR79-promoter;通过双酶切、琼脂糖凝胶电泳、DNA测序和荧光素酶活性测定等实验手段,验证质粒pGL3-WDR79-promoter构建的正确性及其表达活性。本研究结果为进一步探讨人WDR79基因表达的调控机制奠定实验基础。 相似文献

细胞辐射损伤的径迹结构模型给出了V-79中国仓鼠细胞的重离子失活截面与实验相符合的结果,井明显地表现出“径迹宽度区”所特有的thindown现象,计算用的放射灵敏参数是由存活曲线得到的,径向剂量分布用数值积分方法计算。本文的结果与不考虑δ射线发射角的径向剂量分布公式相比,有较大改进,重离子失活戳面与实验符合得更好。 相似文献

Summary Exposure of Chinese hamster cells to pulsing electromagnetic field (PEMF) with 0.18–2.5 mT did not influence the baseline frequency of sister-chromatid exchanges (SCE). The results suggest that PEMF with the magnetic intensity examined does not interfere with DNA replication nor produce DNA lesions, thereby leading to an increased frequency of SCE. 相似文献

Summary DNA synthesis in Chinese hamster V79 cells was significantly enhanced when they were exposed to weak, pulsing electromagnetic fields generated by specific combinations of the pulse width (25s), frequency (10, 100 Hz) and magnetic intensity (2×10–5 8×10–5T).
