Presence of hsp65 in bacterial extracts (OM-89): A possible mediator of orally-induced tolerance?

Heat shock proteins (HSP) have been implicated in rodent models of autoimmunity, particularly arthritis, and there is suggestive though inconclusive evidence that they may also play a role in human autoimmune disease. The simplest hypothesis is based on molecular mimicry due to the amino-acid sequence homology between mammalian and microbial HSP. Recently OM-89, an extract of several strains ofEscherichia coli, has shown some efficacy in the treatment of rheumatoid arthritis (RA) when taken orally. Using species-specific antibodies, we show here that OM-89 contains the 65 kDa HSP (hsp65), while hsp65 was not detected in another bacterial extract containing other microorganisms, includingStaphylococcus aureus (OM-85). We suggest that if the human homologue of hsp65 is a relevant target antigen in the human disease, the efficacy of the preparation could be due to induction of oral tolerance or to switching the Th1 response towards Th2. Alternatively, even if the human hsp65 is not a target molecule in RA joints, OM-89 may evoke bystander suppression of joint inflammation via induction of TGF-secreting effector cells. These hypotheses should be tested in further studies.     

Cyclophosphamide-impaired regulation of hepatic heme metabolism

Summary Im male rats hepatic cytochromes b₅ and P-450 were reduced at different times after treatment with cyclophosphamide (CP) (200 mg/kg i.p. for 3 days). Parallel to the above changes the heme metabolism showed derangement: 65nt543544/xxlarge948.gif" alt="delta" align="BASELINE" BORDER="0">-aminolaevulinate synthase, the rate-limiting enzyme in heme synthesis, was depressed and heme oxygenase, the enzyme which catalyzes the oxidative degradation of heme, was increased.     

X65管线钢焊接接头抗开裂性能及止裂性能

为了研究X65管线钢焊接接头的抗开裂性能和母材的止裂性能，对接头的金相组织进行了观察，并且进行了接头各区域裂纹尖端张开位移(CTOD)试验和母材的落锤撕裂试验(DWTT)。试验结果表明：X65钢焊接接头焊缝和母材相比，组织粗大且不均匀，其冲击韧度和断裂韧度值均明显低于母材；X65钢管材对应剪切面积为40％的温度约为 -35℃，X65钢母材的抗开裂性能明显优于焊缝，且具有良好的止裂性能。     

X65管线钢抗H_2S腐蚀的试验研究

研究了不同显微组织管线钢抗H2S腐蚀的行为·结果表明,酒钢生产的X65管线钢,当显微组织是以针状铁素体为主的混合型组织时,管线钢具有优良的抗H2S腐蚀性能·经硫化氢腐蚀试验后,除出现程度不等的氢鼓泡外,试样断面上基本没有产生裂纹·氢鼓泡是由于钢中夹杂物处吸收由腐蚀而产生的氢所引起的·当钢表面上因腐蚀而释放出原子态氢后,由于硫化氢的催化作用,促进原子氢向钢中扩散,并在夹杂物与基体的界面上聚集形成分子氢·随着过程的进行,产生很高的压力,从而形成鼓泡·     

A 65-nm 1-Gb NOR floating-gate flash memory with less than 50-ns access time

This paper presents a 65-nm 1-Gb NOR-type floating-gate flash memory, in which the cell device and chip circuit are developed and optimized. In order to solve the speed problem of giga-level NOR flash in the deep submicron process, the models of long bit-line and word-line are first given, by which the capacitive and resistive loads could be estimated. Based on that, the read path and key modules are optimized to enhance the chip access property and reliability. With the measurement results, the flash memory cell presents good endurance and retention properties, and the macro is operated with 1-las/byte program speed and less than 50-ns read time under 3.3 V supply.     

基于查找表的AD2S65芯片软件算法实现

由于高性能DRC芯片的禁运,导致货源的紧缺和价格的上扬。为此提出了微处理器控制电路实现DRC芯片功能。介绍了DRC芯片AD2S65的功能特点及工作原理,提出了DRC芯片电路实现的方案——基于C8051F单片机的系统方案,并介绍了硬件结构及软件设计流程。在软件算法实现过程中,为提升数据处理速度,使用查找表的算法,使用最少的器件实现了原芯片的所有功能,并实现了产品模块化、小型化的目的。     

结核分枝杆菌HSP65-IL-2融合基因疫苗的构建及表达

构建结核分枝杆菌HSP65-IL-2融合基因疫苗,并对其在体外真核细胞中进行表达。用EcoRV和Hindm双酶切合HSP65-IL-2融合基因的pPaO-hsp65-IL-2载体,回收目的基因片断HSP65-IL-2,并将其亚克隆入同样双酶切的真核表达载体pcDNA3．1（-）中,重组质粒酶切鉴定正确后以脂质体瞬时转染COS-7细胞,并通过间接免疫荧光技术检测目的蛋白的表达。重组真核表达质粒酶切后可获得约2067bp的融合基因HSP65-IL-2片段,间接免疫荧光检测重组质粒转染的COS-7细胞,可在细胞胞浆中看到特异性的绿色荧光。成功地构建了结核分枝杆菌HSP65-IL-2融合基因的真核表达载体,且该重组载体可在体外真核细胞中获得特异性的表达。

介绍了济钢中厚板厂供国内某管道工程X65管线钢板开发生产情况,采用低C-高Nb—Mn—Ti的经济合金成分设计,配合洁净钢冶炼以及合理的板坯加热、轧制冷却工艺,生产出的X65管线钢板具有较高的强度,良好的低温韧性以及止裂性能。
