Summary The -adrenergic agonist isoproterenol and prostaglandins E1 and E2 (but not F2) increased the cAMP content of rat submandibular acini in vitro, but only isoproterenol enhanced ouabain-sensitive86Rb (K) uptake. These findings suggest that cAMP is not involved in the activation of the Na, K pump in salivary cells.  相似文献

Summary Larvae of the cabbage white butterfly,Pieris brassicae, have a dietary requirement for linolenic acid (C183n3) and were found to accumulate two other members of the n-3 family, C203n3 and C205n3 (eicosapentaenoic acid) especially in testicular phospholipids. Arachidonic acid was observed in trace amounts only. During diapause the relative titer of eicosapentaenoic acid increased in testicular phospholipids to about 4.2% of the fatty acids. Eicosapentaenoic acid is a possible precursor of prostaglandins, suggesting that prostaglandins of the 3-series predominate in this insect.  相似文献

It has been proposed that neuroinflammation, among other factors, may trigger an aberrant neuronal cell cycle re-entry leading to neuronal death. Cell cycle disturbances are also detectable in peripheral cells from Alzheimer’s disease (AD) patients. We previously reported that the anti-inflammatory 15- deoxy-Δ12,14-prostaglandin J 2 (15d-PGJ 2) increased the cellular content of the cyclin-dependent kinase inhibitor p27, in lymphoblasts from AD patients. This work aimed at elucidating the mechanisms of 15d-PGJ 2-induced p27 accumulation. Phosphorylation, half-life, and the nucleo-cytoplasmic traffic of p27 protein were altered by 15d-PGJ2 by mechanisms dependent on PI3K/Akt activity. 15d-PGJ 2 prevents the calmodulin-dependent Akt overactivation in AD lymphoblasts by blocking its binding to the 85-kDa regulatory subunit of PI3K. These effects of 15d-PGJ 2 were not mimicked by 9,10-dihydro-15-deoxy-Δ12,14- prostaglandin J 2, suggesting that 15d-PGJ 2 acts independently of peroxisome proliferator-activated receptor γ activation and that the α,β-unsaturated carbonyl group in the cyclopentenone ring of 15d-PGJ 2 is a requisite for the observed effects. Summary The present work investigates the possibility that lipoxygenase products are involved in the biochemical mechanisms of blastocyst implantation by utilizing nordihydroguaiaretic acid (NDGA) and caffeic acid (CA), inhibitors of lipoxygenase enzymes, and quinacrine (QU), an inhibitor of phospholipase-A2. It has been shown previously that inhibition of cyclooxygenase results in blockade of implantation. The inhibitors were dissolved in a standard medium and 5 l of the solutions were micro-injected into the uterine horns of day-4 pregnant mice. The contralateral horns acted as controls and received only vehicle. A sham-operated group provided normal controls. In 14 NDGA-treated mice, the control horns contained 40 implantations while the treated horns contained only 6 small implantations and 8 resorbing sites. These control horns were comparable to the sham controls. In 14 CA-treated mice, treated horns contained 17 small implantations plus 4 resorptions, whereas the control horns contained 26 small implantations and 4 resorptions. Twelve QU-treated mice exhibited 7 small implantations and 4 resorptions in the treated horns, plus 24 small sites and no resorptions in the control horns. Fourteen sham-operated mice had 95 implantation sites and no resorptions in their 28 horns. The results provide evidence for the involvement of the lipoxygenase enzymes and phospholipase-A2 in the initial implantation process and in the subsequent development of early pregnancy.  相似文献

Summary Frog skeletal muscle incubated in vitro with the ionophore A23187 shows extensive morphological alterations. Myofilament disruption, presumably mediated by excess intracellular calcium, can be partially prevented by preincubating the muscle with inhibitors of prostaglandin synthesis and the lysosomal thiol protease inhibitor leupeptin.The author would like to thank Dr R. Summary Human -atrial natriuretic peptide (h-ANP) makes the urine of dehydrated volunteers hypotonic to plasma despite high circulating concentrations of antidiuretic hormone. Urinary dilution with h-ANP also occurs in subjects receiving indomethacin. Therefore, h-ANP antagonises effects of antidiuretic hormone on distal tubular V2-receptors in man, probably without involving prostaglandins.  相似文献

Summary Restraint-induced stress potentiated morphine catalepsy in rats. This potentiation was partially antagonized by pharmacologic treatments decreasing central serotonin, acetylcholine, prostaglandins and by naloxone. Selective increase in central dopamine also inhibited the potentiation.Acknowledgments.
