CFD-ACE＋软件对PCR微芯片传热过程的数值模拟和分析

针对基于MEMS技术的PCR微芯片温度控制难的问题，通过对该芯片的传热过程进行简化并建立物理模型和数学模型，对芯片内部的热传导和温度变化过程进行了理论推导分析。利用CFD-ACE＋软件对芯片的热传导过程进行了数值模拟。模拟结果表明，该芯片具有较高的升、降温速度。芯片在自行构建的温度控制系统下进行了热循环反应，对GUS基因成功实现了扩增。     

DNA聚合酶链反应鉴测梨树火疫病

对细菌Ｅ．ａｍｙｌｏｖｏｒａ，Ｅ．ｈｅｒｂｉｃｏｌａ和Ｐｓｅｕｄｏｍｏｎａｓｓｙｒｉｎｇａｅ的ＤＮＡ提纯，并通过聚合酶链反应，发现引物Ｂ＿５等可用以有效地鉴别Ｅ．ａｍｙｌｏｖｏｒａ．对Ｅ．ａｍｙｌｏｖｏｒａ细胞培养液直接稀释，加清洁剂处理后进行聚合酶链反应（ＰＣＲ）．据引物Ｂ＿５测定，得到清晰而强烈的相同的ＤＮＡ分子电泳光带，可有效反应测定５００个细菌细胞；用引物Ｂ＿５和Ｂ＿６进一步测定细菌Ｅ．ａｍｙｌｏｖｏｒａ。Ｅ．ｈｅｒｂｉｃｏｌａ和Ｐ．ｓｙｒｉｎｇａｅ，从Ｅ．ｈｅｒｂｉｃｏｄａ反应获得一个电泳光谱，从Ｐ．ｓｙｒｉｎｇａｅ获得３个泳谱；用引物Ｂ＿５和Ｂ＿７与细菌Ｅ．ａｍｙｌｏｖｏｒａ和Ｐ．ｓｙｒｉｎｇａｅ反应，从Ｅ．Ｅ．ａｍｙｌｏｖｏｒａ得到两条相同光带，从Ｐ．ｓｙｒｉｎｇａｅ反应获得了相似的带谱分布。     

在胎儿发育过程中人MK基因的组织差异性表达

MK是肝素结合因子家族中的一个成员，用RT－PCR研究了第11－16周胎儿龄的胎儿的神经组织与非神经组织中MK基因的组织差异性表达，随着胎儿龄的增加，在神经组织中，MK呈现出规律性的从无互有和从低表达到高表达的趋势，提示它可能在胎儿脑的发育中起着重要的作用，在其他非神经组织中，尽管各组织中MK的表达呈现不同的规律，但看起来总的趋势是趋向活跃。     

检验唐氏综合症的法医学方法研究

利用荧光标记的PCR法，对唐氏综合症的检验进行研究．采用法医学检验常用试剂盒AmpFe STR^TM Profiler Plus^TM对唐氏综合症病例进行PCR扩增，扩增产物用ABIPRISMT”310型DNA全自动测序仪分型判断．结果表明，该检验方法具有简便、快速、直接、准确的特点，检验效果非常好，而且可用于产前诊断．该方法用于检验唐氏综合症是一种理想的方法．

The in vivo expression of the globin genes of theβ cistron in γ-,δ-, andδβ-thalassemia heterozygotes

There is considerable evidence suggesting that the switch from to and chain production after birth is due, in part, to silencing of the genes by stage-specific factors which bind to their promoters and to the competition from the adult ( and ) genes for a common enhancer element located in the locus control region. As a consequence one can expect that the increased Hb F production in adults with hereditary persistence of fetal hemoglobin or -thalassemia is directed mainly by -globin genes in cis to the deletion(s) responsible for these conditions. Here we review data on heterozygotes with -, -, or -thalassemia, who also had an^AT mutation, in cis or in trans, which was used as a marker of gene expression. The results show that a deletion affecting adult genes favors the expression of genes in cis, while the deletion of a single gene does not affect the expression of the gene in cis but leads to a faster switch postnatally.     

Weibull分布和极值分布场合下简单步进应力加速寿命试验的最优设计

产品的加速寿命试验被用来较快地获得关于产品寿命分布的信息.本文研究了Weibull分布和极小值分布下简单步加试验的最优设计问题.为解决Nelson折算法对极小值分布的数据处理产生特殊情况的问题,作者应用近年来一些新的研究成果,将位置-尺度分布中的尺度参数设为随应力变化的量进行最优设计.给出一实例,并将其推广到更一般的形式.     

Cloning and expression of the vp39 gene ofBombyx mori nuclear polyhedrosis virus inE. coli

The nuclear capsid protein gene (vp39) ofBombyx mori nuclear polyhedrosis virus (BmNPV) was amplified successfully by PCR technique and inserted into pGEM 3zf(+). The 5′ and 3′ terminal area of the amplified vp39 gene were sequenced with silver-staining dideoxy method. Bmvp39 gene was sub-cloned into the expression vector pRSET-A, and transformed intoE. coli BL21. This gene was highly expressed by IPTG induction. SDS-PAGE analysis showed that the expressed protein is about 38 kd, and the expressed amount reached maxium in 4 h with IPTG induction. Rh—Ag／SiO2催化剂上CO的吸附与加氢反应

应用表面反应红外动态技术考察了促进型铑催化剂Ｒｈ－Ａｇ／ＳｉＯ２上ＣＯ吸附及其加氢反应的性能,结果表明：线式吸附态的ＣＯ对氢的反应性能高于桥式吸附的ＣＯ,是加氢反应的主要活性吸附态．反应条件下温度升高有利于Ｒｈ－Ａｇ／ＳｉＯ２上线／桥ＣＯ吸附态强度比值的增加．表面反应生成了表面甲酰基、甲酸盐、乙酰基、乙酸盐、乙烯酮及烯醇盐等关键的Ｃ１－２含氧中间体．助剂Ａｇ的加入提高了铑催化剂上ＣＯ吸附态的线／桥强度比值以及线式吸附ＣＯ的加氢反应速率和Ｃ２含氧物的生成率．
