Damage to DNA in the cell activates the tumour-suppressor protein p53, and failure of this activation leads to genetic instability and a predisposition to cancer. It is therefore crucial to understand the signal transduction mechanisms that connect DNA damage with p53 activation. The enzyme known as DNA-dependent protein kinase (DNA-PK) has been proposed to be an essential activator of p53, but the evidence for its involvement in this pathway is controversial. We now show that the p53 response is fully functional in primary mouse embryonic fibroblasts lacking DNA-PK: irradiation-induced DNA damage in these defective fibroblasts induces a normal response of p53 accumulation, phosphorylation of a p53 serine residue at position 15, nuclear localization and binding to DNA of p53. The upregulation of p53-target genes and cell-cycle arrest also occur normally. The DNA-PK-deficient cell line SCGR11 contains a homozygous mutation in the DNA-binding domain of p53, which may explain the defective response by p53 reported in this line. Our results indicate that DNA-PK activity is not required for cells to mount a p53-dependent response to DNA damage.  相似文献

HUMAN fibroblasts in culture synthesise both type I and type III collagen(1), with type I accounting for 70-90% of the total(2). In culture, the rates at which these proteins are synthesised is constant and apparently rather rigidly controlled(3). However, the proportions of these collagens differs in cells cultured with increased amounts of serum (increased type III/I)(4) as well as in cells obtained from patients with certain diseases. Cells from patients with the Ehlers-Danlos type IV syndrome make little or no type III collagen(5,6), whereas cells from patients with osteogenesis imperfecta have an increased type III/I (refs 7, 8). We have found that cells from some patients with systemic sclerosis (scleroderma), have a reduced type III/I ratio. However, as previously reported, these cells grew to a lower density than control cells(9). We report here that normal fibroblasts from human and guinea pig skin produce proportionally more type III collagen at high cell density, probably because of a reduction in the synthesis of type I collagen.  相似文献

Fullerenes are graphitic cage structures incorporating exactly twelve pentagons. The smallest possible fullerene is thus C20, which consists solely of pentagons. But the extreme curvature and reactivity of this structure have led to doubts about its existence and stability. Although theoretical calculations have identified, besides this cage, a bowl and a monocyclic ring isomer as low-energy members of the C20 cluster family, only ring isomers of C20 have been observed so far. Here we show that the cage-structured fullerene C20 can be produced from its perhydrogenated form (dodecahedrane C20H20) by replacing the hydrogen atoms with relatively weakly bound bromine atoms, followed by gas-phase debromination. For comparison we have also produced the bowl isomer of C20 using the same procedure. We characterize the generated C20 clusters using mass-selective anion photoelectron spectroscopy; the observed electron affinities and vibrational structures of these two C20 isomers differ significantly from each other, as well as from those of the known monocyclic isomer. We expect that these unique C20 species will serve as a benchmark test for further theoretical studies.  相似文献

采用文献计量学方法,对《华南理工大学学报(自然科学版)》创刊以来出版的315期的载文、栏目、作者和引文进行统计分析,并与相关的文献报道进行比较.结果表明:1995年(月刊)以来,年载文量基本上稳定在300篇左右,论文发表时滞逐渐缩短.基金论文比逐年提高,近年来稳定在90%以上.“电子、通信与自动控制”栏目的载文量最多,与“计算器科学与技术”、“机械工程”、“化学、化工(能源)”、“土木建筑工程”等5个栏目是该刊的主栏目.校内作者是该刊的第一大作者群,其次为其他高校的作者,二者合计超过作者总数的95%;第一作者中具有高级职称的比例在40%左右,近年来博士后、博士和博士研究生的比例高达70%左右,作者队伍优秀.该刊的引文数量多、种类多样、质量上乘;2012年的载文中,篇均引文为16.99条,英文引文占68.64%,期刊类引文占74.37%,单篇引文最多的达84条,最少的有8条.  相似文献
