Peroxidase activity of neutrophil granules in two cases of congenital myeloperoxidase deficiency]

The neutrophils and monocytes of two patients with hereditary myeloperoxidase deficiency lacked myeloperoxidase activity as determined by light and electron microscopic cytochemical staining. Using Graham-Karnovsky media, azurophils of neutrophils were devoid of peroxidase whereas all eosinophilic and basophilic granules exhibited normal peroxidase activity. After incubation in alkaline diaminobenzidine media which stains the catalase of microperoxisomes, some small granules were seen to be strongly stained in both immature and mature neutrophils. These small granules were distinct from all other neutrophilic granules which lacked a positive reaction. Only, in the presence of cyanide or aminotriazole, peroxidatic activity was also detected in some ellipsoid azurophils. This observation suggests that these substances activated an oxidase whose nature is discussed.     

Millennial-scale paleoceanography in Okinawa Trough during Late Quaternary period

Based on the analyses of foraminifer and accelerator mass spectrometer radiocarbon dating in DGKS9603 core from mid-Okinawa Trough close to bottom, oscillation curve, which expressed the relation between the surface water temperature and the depth, has been obtained by using foraminifer analysis and calculation of FP-12E transfer function. The whole core indicated seven cold phases and eight warm phases. Obvious expression of low temperature event during Middle and Late Holocene, YD,H1,H2,H3 and H4 events, as well as the short cold phase during the middle last glacial period, implied that short shifts since 50 kaBP would have been global significance. Sedimentation rate during cold phases is usually faster than that in warm stages, with the lowest rate in Holocene, which may be connected with rising sea level and principal axial of Kuroshio Current moving to west. Volcanic activities highly developed in Okinawa Trough during the Quaternary period, thus abundant volcanic glass and pumice were well preserved.

Folliculo-stellar cells are present in the anterior lobe of hypophysis of the monkey Macacus irus. These cells limit strait cavities in which one can observe microvillosites at the apical pole of the cells. The folliculo-stellar cells possess long and thin fibers which insert themselves between different types of granular cells and sometimes extend to the peri-capillar regions. Numerous organelles are present in these folliculo-stellar cells, especially microfilaments.

Anthrax toxins cooperatively inhibit endocytic recycling by the Rab11/Sec15 exocyst

Bacillus anthracis is the causative agent of anthrax in humans and other mammals. In lethal systemic anthrax, proliferating bacilli secrete large quantities of the toxins lethal factor (LF) and oedema factor (EF), leading to widespread vascular leakage and shock. Whereas host targets of LF (mitogen-activated protein-kinase kinases) and EF (cAMP-dependent processes) have been implicated in the initial phase of anthrax, less is understood about toxin action during the final stage of infection. Here we use Drosophila melanogaster to identify the Rab11/Sec15 exocyst, which acts at the last step of endocytic recycling, as a novel target of both EF and LF. EF reduces levels of apically localized Rab11 and indirectly blocks vesicle formation by its binding partner and effector Sec15 (Sec15-GFP), whereas LF acts more directly to reduce Sec15-GFP vesicles. Convergent effects of EF and LF on Rab11/Sec15 inhibit expression of and signalling by the Notch ligand Delta and reduce DE-cadherin levels at adherens junctions. In human endothelial cells, the two toxins act in a conserved fashion to block formation of Sec15 vesicles, inhibit Notch signalling, and reduce cadherin expression at adherens junctions. This coordinated disruption of the Rab11/Sec15 exocyst by anthrax toxins may contribute to toxin-dependent barrier disruption and vascular dysfunction during B. anthracis infection.
