Mouse knockout technology provides a powerful means of elucidating gene function in vivo, and a publicly available genome-wide collection of mouse knockouts would be significantly enabling for biomedical discovery. To date, published knockouts exist for only about 10% of mouse genes. Furthermore, many of these are limited in utility because they have not been made or phenotyped in standardized ways, and many are not freely available to researchers. It is time to harness new technologies and efficiencies of production to mount a high-throughput international effort to produce and phenotype knockouts for all mouse genes, and place these resources into the public domain.  相似文献

The evolution of the multiple perspective approach and its range of applications over the past decade are reviewed. The traditional technical perspective of systems analysis is augmented with organizational and personal perspectives. The three types of perspectives have inherently different characteristics and properties. The applications show that each perspective yields insights on a system that are not attainable with the others. The organizational and personal perspectives also focus more attention on problems of implementation. The concept is serving as an effective and practical vehicle to overcome the limitations of systems analysis in dealing with complex real-world situations.  相似文献

Proviruses of the mouse mammary tumour virus (MMTV) endogenous to normal mice can be identified by molecular hybridisation and distinguished using restriction endonucleases. Feral mice display marked heterogeneity with respect to the number of copies and the sites of insertion of endogenous MMTV-specific DNA, with occasional mice apparently free of MMTV DNA. Several different MMTV proviruses present in laboratory mice have segregated like stable, independent genetic elements during the inbreeding which followed a cross between Bagg albino and DBA mice 60 years ago. The results favour the hypothesis that endogenous proviruses have been established by multiple, independent infections of germ cells rather than by somatic mutation of ancestral proviruses or of cellular genes.  相似文献

Electron-electron interactions can render an otherwise conducting material insulating, with the insulator-metal phase transition in correlated-electron materials being the canonical macroscopic manifestation of the competition between charge-carrier itinerancy and localization. The transition can arise from underlying microscopic interactions among the charge, lattice, orbital and spin degrees of freedom, the complexity of which leads to multiple phase-transition pathways. For example, in many transition metal oxides, the insulator-metal transition has been achieved with external stimuli, including temperature, light, electric field, mechanical strain or magnetic field. Vanadium dioxide is particularly intriguing because both the lattice and on-site Coulomb repulsion contribute to the insulator-to-metal transition at 340?K (ref. 8). Thus, although the precise microscopic origin of the phase transition remains elusive, vanadium dioxide serves as a testbed for correlated-electron phase-transition dynamics. Here we report the observation of an insulator-metal transition in vanadium dioxide induced by a terahertz electric field. This is achieved using metamaterial-enhanced picosecond, high-field terahertz pulses to reduce the Coulomb-induced potential barrier for carrier transport. A nonlinear metamaterial response is observed through the phase transition, demonstrating that high-field terahertz pulses provide alternative pathways to induce collective electronic and structural rearrangements. The metamaterial resonators play a dual role, providing sub-wavelength field enhancement that locally drives the nonlinear response, and global sensitivity to the local changes, thereby enabling macroscopic observation of the dynamics. This methodology provides a powerful platform to investigate low-energy dynamics in condensed matter and, further, demonstrates that integration of metamaterials with complex matter is a viable pathway to realize functional nonlinear electromagnetic composites.  相似文献

A Luttinger liquid is an interacting one-dimensional electronic system, quite distinct from the 'conventional' Fermi liquids formed by interacting electrons in two and three dimensions. Some of the most striking properties of Luttinger liquids are revealed in the process of electron tunnelling. For example, as a function of the applied bias voltage or temperature, the tunnelling current exhibits a non-trivial power-law suppression. (There is no such suppression in a conventional Fermi liquid.) Here, using a carbon nanotube connected to resistive leads, we create a system that emulates tunnelling in a Luttinger liquid, by controlling the interaction of the tunnelling electron with its environment. We further replace a single tunnelling barrier with a double-barrier, resonant-level structure and investigate resonant tunnelling between Luttinger liquids. At low temperatures, we observe perfect transparency of the resonant level embedded in the interacting environment, and the width of the resonance tends to zero. We argue that this behaviour results from many-body physics of interacting electrons, and signals the presence of a quantum phase transition. Given that many parameters, including the interaction strength, can be precisely controlled in our samples, this is an attractive model system for studying quantum critical phenomena in general, with wide-reaching implications for understanding quantum phase transitions in more complex systems, such as cold atoms and strongly correlated bulk materials.  相似文献
