The cystic fibrosis transmembrane conductance regulator (CFTR) is responsible for the disease cystic fibrosis (CF). It is a membrane protein belonging to the ABC transporter family functioning as a chloride/anion channel in epithelial cells around the body. There are over 1500 mutations that have been characterised as CF-causing; the most common of these, accounting for ~70 % of CF cases, is the deletion of a phenylalanine at position 508. This leads to instability of the nascent protein and the modified structure is recognised and then degraded by the ER quality control mechanism. However, even pharmacologically ‘rescued’ F508del CFTR displays instability at the cell’s surface, losing its channel function rapidly and it is rapidly removed from the plasma membrane for lysosomal degradation. This review will, therefore, explore the link between stability and structure/function relationships of membrane proteins and CFTR in particular and how approaches to study CFTR structure depend on its stability. We will also review the application of a fluorescence labelling method for the assessment of the thermostability and the tertiary structure of CFTR.  相似文献

Home range size and foraging habitat use in breeding lesser kestrels (Falco naumanni), a bird species of conservation concern, were investigated during the breeding season of the species in 2008 in an intensively cultivated area of central Greece, using radio-tracking. Grasshopper (the main prey) densities were measured at the most important habitats (cotton, cereals, grasslands and margins). Home ranges were not significantly different between sexes either as overall means or during incubation and nestling periods. Movements of both sexes were non-random during incubation but random during the nestling period. Habitats used by males during incubation ranked as: margins > other > cotton > corn > cereals and during nestling period as: cereals > margins > grasslands > corn > cotton. Female habitat use greatly differed ranking as cereals > cotton > grasslands during incubation and as grassland > cotton > corn > cereals > margins during nestling period. Female habitat use seemed to be in disagreement with the conditions generally favouring prey availability, probably for reasons associated with low and uniform distribution of grasshopper densities over the habitats.  相似文献

Lagocephalus sceleratus, is a Lessepsian invasive fish species introduced into the Mediterranean Sea through the Suez Canal. This study investigated it as host of parasites. Out of 41 individuals caught from northern and southern locations of the eastern Aegean Sea, nine were found to host parasites. Gnathia sp. praniza larva, an ectoparasite, was associated with severe damage of the primary and secondary structures of the gills and it was found in one specimen (prevalence 2.4%). Regarding endoparasites, Anisakis type I third-stage larvae were found in two specimens (prevalence 4.9%). In addition, adult nematodes of Hysterothylacium aduncum were found in another six specimens (prevalence 14.6%). Lagocephalus sceleratus is a new host for some indigenous parasites, with low host specificity, in the Mediterranean Sea, providing an additional niche for the success and increase of local populations of these parasites and, to our knowledge, this is the first record of L. sceleratus parasites in this area. Whether these parasites can control the invasion success of L. sceleratus needs to be further investigated. However, the low mean intensity of infection found in this study does not support such control.  相似文献
