By utilizing the air quality monitoring data and the NCEP reanalysis data, the relationship between the PM10 air pollution episode and synoptic situation is analyzed in the Beijing area. It is interesting to find that PM10 air pollution episode in and around the Beijing area is correlated with the Western Pacific tropical cyclone track to some extent, namely when a tropical cyclone lands southward to the Changjiang River, PM10 air pollution episode is not easy to take place generally in the Beijing area; but when a tropical cyclone moves northward and finally lands at the Korea Peninsula or the Japanese mainland, and under this condition the Beijing area is generally controlled by weak high or weak low for several days, PM10 air pollution episode often takes place in one day or several days. Above findings indicate that predicting the Western Pacific tropical cyclone track can foretell whether or not PM10 air pollution episode takes place in the Beijing area, which can offer a technique for urban air quality prediction and air pollution source management in the Beijing area. 锌指核酸酶介导的高效多位点基因打靶   总被引:1,自引:0,他引:1  
该研究旨在建立锌指核酸酶介导的多位点基因打靶技术,为获得稳定遗传的转基因动物或基因治疗临床应用解决技术难题.首先利用OPEN平台设计、构建能识别人基因组内rDNA基因间隔序列的锌指蛋白基因序列,与FokⅠ的切割结构域连接、表达后获得锌指核酸酶基因,再构建锌指核酸酶真核表达载体.另外,构建含有2条同源重组引导序列和绿色荧光蛋白基因(EGFP)的多位点基因打靶载体.将锌指核酸酶真核表达载体和多位点基因打靶载体共转染HEK293细胞,内参对照PCR-灰度分析法检测外源基因定点整合效率,结果显示单独转染多位点基因打靶载体的定点整合效率为6.8%;而由于锌指核酸酶在染色质DNA上切割rDNA基因的间隔序列,诱导高效同源重组,锌指核酸酶载体、多位点基因打靶载体共转染的定点整合效率为24.2%,较常规基因打靶定点整合率(10-6~10-5)提高了24000多倍.共转染的HEK293细胞在无任何筛选的条件下持续培养2个月,经过20次传代之后,子代细胞能够持续表达EGFP,提示表达稳定.本研究建立了锌指核酸酶介导的高效多位点基因打靶技术,不仅大大提高了外源基因的定点整合效率,而且兼顾了基因表达的稳定性和安全性,为动物定点转基因和人类基因治疗提供了重要的技术平台,具有广泛的应用前景.  相似文献

This paper studies interdecadal variations of heat waves and sultry weather (HW&SW) in Beijing City by using climatological data gathered from 1940-2000.In general, HW&SW decreased during the 1940s-1970s and increased during the 1970s-1990s.Although variation trends in the number of HW&SW days coincide with air temperature variations, the greatest number of HW&SW days occurred during the 1940s rather than during the much hotter 1990s (which had an average 1.0°C higher air tem-perature than during the 1940s)...  相似文献

以1,6-二甲氧基萘为原料,经Na/C2H5OH还原生成5-甲氧基-2-四氢萘酮,与正丙胺还原胺化生成外消旋化合物2-(N-正丙基氨基)-5-甲氧基四氢化萘,经拆分得到手性的化合物(-)-2-(N-正丙基氨基)-5-甲氧基四氢化萘,再去甲基反应得到化合物(-)-2-(N-正丙基氨基)-5-羟基四氢化萘。采用元素分析、红外光谱、1H-NMR和13C-NMR表征了中间体和产物的分子结构。通过合成工艺的研究,得到较好的工艺条件,反应总收率达16.8%。采用一步法的还原胺化反应,不仅简化了反应步骤,而且收率提高了16.2%。  相似文献
