Intestinal morphogenesis and differentiation are dependent on heterotypic cell interactions between embryonic epithelial cells (endoderm) and stromal cells (mesenchyme). Extracellular matrix molecules represent attractive candidates for regulators of these interactions. The structural and functional diversity of the extracellular matrix as intestinal development proceeds is demonstrated by 1) spatio-temporal specific expression of the classically described constituents, 2) the finding of laminin and collagen IV variants, 3) changes in the ratio of individual constituent chains, and 4) a stage-specific regulation of basement membrane molecule production, in particular by glucocorticoids. The orientation/assembly of these extracellular matrix molecules could direct precise cellular functions through interactions via integrin molecules. The involvement of extracellular matrix, and in particular basement membrane molecules in heterotypic cell interactions leading to epithelial cell differentiation, has been highlighted by the use of experimental models such as cocultures, hybrid intestines and antisense approaches. These models allowed us to conclude that a correct elaboration and assembly of the basement membrane, following close contacts between epithelial and fibroblastic cells, is necessary for the expression of differentiation markers such as digestive enzymes. 相似文献
This note presents a new result of terrestrial mollusk study from the Luochuan loess section since the last 250 ka. A total
of 213 samples, taken at intervals of 10 cm in the S0-L3 portion, were analyzed for fossil mollusks. Generally, 150–600 individuals were counted in each sample. According to the
distribution of mollusk fossil assemblages in the loess section, 11 mollusk fossil zones have been recognized, representing
different climatic and ecological conditions. Three main ecological groups were identified according to the temperature and
moisture requirements of each taxon. The cold-aridiphilous group shows maxima at about 240–220, 190–182, 150–140 and 74–66
ka BP. The thermo-humidiphilous set has high abundances for at least 6 times in the section at about 246–240, 220–216, 170–158,
92–86, 60–44 and 10 kaBP. Our results show that variations in mollusk ecological groups are related with changes in the Earth
orbital parameters at the 41 and 20 ka frequencies. Maxima in thermo-humidiphilous taxa are in phase with accretion intervals
of obliquity (41 ka period), reflecting the East Asian summer monsoon with 41 ka period in controlling variations in terrestrial
mollusk ecological groups. In addition, maxima in thermohumidiphilous taxa appearing at about 246–240, 220–216, 60 and 10
kaBP, are consistent with procession (20 ka period) maxima, indicating that the procession period also plays an important
role in adjusting the ecological pattern of mollusk groups. 相似文献
In this article we discuss how an interdisciplinary research team partnered with a variety of stakeholders concerned with and/or affected by the impacts of climate change in the Red River Delta of Vietnam. The research, undertaken from 2016 to 2018, drew upon a wide range of methods to investigate systemically these impacts – with a view to the research inputting into the development of (more) sustainable ways of living. The research solicited various accounts of the experience of climate change in the community, set up learning processes in community meetings, and created an interface with government officials positioned at commune, district, provincial, and national levels. The intention was to offer support towards developing a learning process (broadly defined as including learnings/systemic inquiry across organizational levels of the society) to pursue options for sustainable living. The article offers our post-facto reflections which render more explicit (to ourselves and for the benefit of audiences) how the research team, with Hoang as lead researcher, facilitated the inquiry process towards developing a synthesis which underscored the assets for resilience to climate change and supported interventions to strengthen such (defined) assets.
Particle accelerators are used in a wide variety of fields, ranging from medicine and biology to high-energy physics. The accelerating fields in conventional accelerators are limited to a few tens of MeV m(-1), owing to material breakdown at the walls of the structure. Thus, the production of energetic particle beams currently requires large-scale accelerators and expensive infrastructures. Laser-plasma accelerators have been proposed as a next generation of compact accelerators because of the huge electric fields they can sustain (>100 GeV m(-1)). However, it has been difficult to use them efficiently for applications because they have produced poor-quality particle beams with large energy spreads, owing to a randomization of electrons in phase space. Here we demonstrate that this randomization can be suppressed and that the quality of the electron beams can be dramatically enhanced. Within a length of 3 mm, the laser drives a plasma bubble that traps and accelerates plasma electrons. The resulting electron beam is extremely collimated and quasi-monoenergetic, with a high charge of 0.5 nC at 170 MeV. 相似文献
Synchrotrons have for decades provided invaluable sources of soft X-rays, the application of which has led to significant progress in many areas of science and technology. But future applications of soft X-rays--in structural biology, for example--anticipate the need for pulses with much shorter duration (femtoseconds) and much higher energy (millijoules) than those delivered by synchrotrons. Soft X-ray free-electron lasers should fulfil these requirements but will be limited in number; the pressure on beamtime is therefore likely to be considerable. Laser-driven soft X-ray sources offer a comparatively inexpensive and widely available alternative, but have encountered practical bottlenecks in the quest for high intensities. Here we establish and characterize a soft X-ray laser chain that shows how these bottlenecks can in principle be overcome. By combining the high optical quality available from high-harmonic laser sources (as a seed beam) with a highly energetic soft X-ray laser plasma amplifier, we produce a tabletop soft X-ray femtosecond laser operating at 10 Hz and exhibiting full saturation, high energy, high coherence and full polarization. Cytochrome c oxidase catalyses the 4-electron reduction of dioxygen to water and translocates protons vectorially across the inner mitochondrial membrane. Proposed reaction pathways for the catalytic cycle of the O2 reduction are difficult to verify without knowing the structures of the intermediates, but we now have such information for the catalytic intermediates in the first steps of the reaction of O2 with cytochrome c oxidase from resonance Raman spectroscopy, a technique that enables iron-ligand stretching modes to be identified. Here we report on two more key intermediates: a ferryl-oxo (Fe4 = O2-) and a ferric-hydroxy (Fe3+--OH-) intermediate at the level of 3- and 4-electron reduction, respectively. We identified these intermediates by their characteristic iron-oxygen stretching frequencies (786 cm-1 for Fe4+ = O2-, and 450 cm-1 for Fe3+ -- OH-) and oxygen and deuterium isotope shifts. The oxo atom in the ferryl intermediate is hydrogen-bonded and the iron-oxygen bond in the hydroxy intermediate is anomalously weak. With the identification of the primary, ferryl and hydroxy intermediates, the predominant structures at almost all stages of O2 reduction are now known and the catalytic pathway can be described with more certainty. 相似文献
