Genetic susceptibility to multiple sclerosis is associated with genes of the major histocompatibility complex (MHC), particularly HLA-DRB1 and HLA-DQB1 (ref. 1). Both locus and allelic heterogeneity have been reported in this genomic region. To clarify whether HLA-DRB1 itself, nearby genes in the region encoding the MHC or combinations of these loci underlie susceptibility to multiple sclerosis, we genotyped 1,185 Canadian and Finnish families with multiple sclerosis (n = 4,203 individuals) with a high-density SNP panel spanning the genes encoding the MHC and flanking genomic regions. Strong associations in Canadian and Finnish samples were observed with blocks in the HLA class II genomic region (P < 4.9 x 10(-13) and P < 2.0 x 10(-16), respectively), but the strongest association was with HLA-DRB1 (P < 4.4 x 10(-17)). Conditioning on either HLA-DRB1 or the most significant HLA class II haplotype block found no additional block or SNP association independent of the HLA class II genomic region. This study therefore indicates that MHC-associated susceptibility to multiple sclerosis is determined by HLA class II alleles, their interactions and closely neighboring variants.  相似文献

Effects of the weld microstructure and inclusions on brittle fracture initiation are investigated in a thermally aged ferritic high-nickel weld of a reactor pressure vessel head from a decommissioned nuclear power plant.As-welded and reheated regions mainly consist of acicular and polygonal ferrite,respectively.Fractographic examination of Charpy V-notch impact toughness specimens reveals large inclusions(0.5-2.5μm)at the brittle fracture primary initiation sites.High impact energies were measured for the specimens in which brittle fracture was initiated from a small inclusion or an inclusion away from the V-notch.The density,geometry,and chemical composition of the primary initiation inclusions were investigated.A brittle fracture crack initiates as a microcrack either within the multiphase oxide inclusions or from the debonded interfaces between the uncracked inclusions and weld metal matrix.Primary fracture sites can be determined in all the specimens tested in the lower part of the transition curve at and below the 41-J reference impact toughness energy but not above the mentioned value because of the changes in the fracture mechanism and resulting changes in the fracture appearance.  相似文献

The seismic design and analysis of nuclear power plant (NPP) begin with the seismic hazard assessment and design ground motion development for the site. The following steps are needed for the seismic hazard assessment and design ground motion development: a. the development of regional seismo-tectonic model with seismic source areas within 500 km radius centered to the site; b. the development of strong motion prediction equations; c. logic three development for taking into account uncertainties and seismic hazard quantification; d. the development of uniform hazard response spectra for ground motion at the site; e. simulation of acceleration time histories compatible with uniform hazard response spectra. The following phase two in seismic design of NPP structures is the analysis of structural response for the design ground motion. This second phase of the process consists of the following steps: a. development of structural models of the plant buildings; b. development of the soil model underneath the plant buildings for soil-structure interaction response analysis; c. determination of in-structure response spectra for the plant buildings for the equipment response analysis. In the third phase of the seismic design and analysis the equipment is analyzed on the basis of in-structure response spectra. For this purpose the structural models of the mechanical components and piping in the plant are set up. In large 3D-structural models used today the heaviest equipment of the primary coolant circuit is included in the structural model of the reactor building. In the fourth phase the electrical equipment and automation and control equipment are seismically qualified with the aid of the in-structure spectra developed in the phase two using large three-axial shaking tables. For this purpose the smoothed envelope spectra for calculated in-structure spectra are constructed and acceleration time is fitted to these smoothed envelope spectra.  相似文献

Elements in the alkali metal series are regarded as unlikely superconductors because of their monovalent character. A superconducting transition temperature as high as 20 K, recently found in compressed lithium (the lightest alkali element), probably arises from pressure-induced changes in the conduction-electron band structure. Superconductivity at ambient pressure in lithium has hitherto remained unresolved, both theoretically and experimentally. Here we demonstrate that lithium is a superconductor at ambient pressure with a transition temperature of 0.4 mK. As lithium has a particularly simple conduction electron system, it represents an important case for any attempts to classify superconductors and transition temperatures, especially to determine if any non-magnetic configuration can exclude superconductivity down to zero temperature. Furthermore, the combination of extremely weak superconductivity and relatively strong nuclear magnetism in lithium would clearly lead to mutual competition between these two ordering phenomena under suitably prepared conditions.  相似文献
