Summary Transients in myoplasmic [Ca2+] and in phosphorylation of the 20,000 dalton light chain of myosin have been reported following stimulation of vascular smooth muscle by various agonists. Since these transients are rapid compared with the time required to attain a steady-state stress, agonist diffusion rates may be a significant limitation in activation. The purpose of this study was to estimate the effect of agonist diffusion rates on the time course of activation as assessed by mechanical measurements of stress development and isotonic shortening velocities and by determinations of the time course of myosin phosphorylation. The approach was to measure these parameters in K+-stimulated preparations of the swine carotid media of varying thicknesses and to estimate the theoretical contributions imposed by diffusion rates and the presence of a diffusion boundary layer surrounding the tissue. The results show that the time course of parameters which are tissue averages such as stiffness, active stress, and myosin phosphorylation is dominated by agonist diffusion rates. The sequence of events involved in excitation-contraction coupling including agonist actions on the cell membrane, Ca2+ release, activation of myosin light chain kinase, and cross-bridge phosphorylation appear to be very rapid events compared with stress development. Estimates of unloaded or lightly loaded shortening velocities which are not simple tissue averages appear to provide an imporoved estimate of activation rates.Supported by a grant from the National Institutes of Health 5-PO1-HL19242. Based on the study on the city transport systems of some typical cities worldwide, this paper put forward that each city transport system has its own development mode, which is influenced by the city development plan, economic development level, traveling vehicle composition etc.. When some problems occur, such as the congestions caused by contradiction between the road capacity and vehicle composition, the city transport system may come into temporary maturity period. If the improvement for road system is limited meanwhile, optimized structure of vehicle composition should be an effective solution in this case. With the development of economy-internationalization, the development speed of city transport modernization is rapid. When traveling easiness is conflicting with efficiency, the advantages of public transport system become more obvious. Correspondingly, the superiority of two-wheel vehicles will reappear. The purpose of this paper is to examine the possibilities for extending organisational metaphors into the area of information systems. The mechanistic metaphor is used as a relevant example because analysing the assumptions behind it offers a particularly revealing insight into common assumptions in the field of management studies and information systems. These assumptions were developed in a specific historical context, that of the development of bureaucratic forms in a government and industry and the acceptance of what Weber described as instrumental rationality. Three elements of a characteristically mechanistic view of information management are identified: corporate ownership of information, a formal division of information work and a definition of the nature of information as data whose meaning is noncontroversial. It is argued that this view of information is too restricted to allow for creative development of information use, but that the corporate ownership of information continues to be relevant because of the notion of formal rights which has been developed from it.  相似文献

This paper introduces a non-assembly manufacturing case with microstereolithography technology. The design and manufacturing process of a pneumatic thrust bearing is described, and a special tessellation method is developed to further improve the capability of the manufacturing system thus bigger products can also be easily manufactured. Implemented in a layer-by-layer fashion, stereolithography has been used for the rapid manufacturing of complex devices, and it avoids the expensive assembly process in the traditional manufacturing. This paper presents that microstereolithography can produce high-resolution products with intricate details, small openings, and smooth surfaces. The potential of the microstereolithograhy technique is explored for the rapid manufacturing of small and complex objects.  相似文献

Physical forces elicit biochemical signalling in a diverse array of cells, tissues and organisms, helping to govern fundamental biological processes. Several hypotheses have been advanced that link physical forces to intracellular signalling pathways, but in many cases the molecular mechanisms of mechanotransduction remain elusive. Here we find that compressive stress shrinks the lateral intercellular space surrounding epithelial cells, and triggers cellular signalling via autocrine binding of epidermal growth factor family ligands to the epidermal growth factor receptor. Mathematical analysis predicts that constant rate shedding of autocrine ligands into a collapsing lateral intercellular space leads to increased local ligand concentrations that are sufficient to account for the observed receptor signalling; direct experimental comparison of signalling stimulated by compressive stress versus exogenous soluble ligand supports this prediction. These findings establish a mechanism by which mechanotransduction arises from an autocrine ligand-receptor circuit operating in a dynamically regulated extracellular volume, not requiring induction of force-dependent biochemical processes within the cell or cell membrane.  相似文献
