Endomembranes of eukaryotic cells are dynamic structures that are in continuous communication through the activity of specialized cellular machineries, such as the coat protein complex II (COPII), which mediates cargo export from the endoplasmic reticulum (ER). COPII consists of the Sar1 GTPase, Sec23 and Sec24 (Sec23/24), where Sec23 is a Sar1-specific GTPase-activating protein and Sec24 functions in cargo selection, and Sec13 and Sec31 (Sec13/31), which has a structural role. Whereas recent results have shown that Sec23/24 and Sec13/31 can self-assemble to form COPII cage-like particles, we now show that Sec13/31 can self-assemble to form minimal cages in the absence of Sec23/24. We present a three-dimensional reconstruction of these Sec13/31 cages at 30 A resolution using cryo-electron microscopy and single particle analysis. These results reveal a novel cuboctahedron geometry with the potential to form a flexible lattice and to generate a diverse range of containers. Our data are consistent with a model for COPII coat complex assembly in which Sec23/24 has a non-structural role as a multivalent ligand localizing the self-assembly of Sec13/31 to form a cage lattice driving ER cargo export.  相似文献

Amorphous materials are commonly understood to consist of random organizations of molecular-type structural units. However, it has long been known that structural organizations intermediate between discrete chemical bonds and periodic crystalline lattices are present even in liquids. Numerous models--including random networks and crystalline-type structures with networks composed of clusters and voids--have been proposed to account for this intermediate-range order. Nevertheless, understanding and controlling structural features that determine intermediate-range order in amorphous materials remain fundamental, yet presently unresolved, issues. The most characteristic signature of such order is the first peak in the total structure factor, referred to as the first sharp diffraction peak or 'low Q' structure. These features correspond to large real-space distances in the materials, and understanding their origin is key to unravelling details of intermediate-range order. Here we employ principles of crystal engineering to design specific patterns of intermediate-range order within amorphous zinc-chloride networks. Using crystalline models, we demonstrate the impact of various structural features on diffraction at low values of Q. Such amorphous network engineering is anticipated to provide the structure/property relationships necessary to tailor specific optical, electronic and mechanical properties.  相似文献

Localization to Xq27 of a susceptibility gene for testicular germ-cell tumours   总被引:13,自引:0,他引:13  
Testicular germ-cell tumours (TGCT) affect 1 in 500 men and are the most common cancer in males aged 15-40 in Western European populations. The incidence of TGCT has risen dramatically over the last century. Known risk factors for TGCT include a history of undescended testis (UDT), testicular dysgenesis, infertility, previously diagnosed TGCT (ref. 7) and a family history of the disease. Brothers of men with TGCT have an 8-10-fold risk of developing TGCT (refs 8,9), whereas the relative risk to fathers and sons is fourfold (ref. 9). This familial relative risk is much higher than that for most other types of cancer. We have collected samples from 134 families with two or more cases of TGCT, 87 of which are affected sibpairs. A genome-wide linkage search yielded a heterogeneity lod (hlod) score of 2.01 on chromosome Xq27 using all families compatible with X inheritance. We obtained a hlod score of 4.7 from families with at least one bilateral case, corresponding to a genome-wide significance level of P=0.034. The proportion of families with UDT linked to this locus was 73% compared with 26% of families without UDT (P=0.03). Our results provide evidence for a TGCT susceptibility gene on chromosome Xq27 that may also predispose to UDT.  相似文献

In this article we reflect upon an integrated approach to action research. The role of the engaged researcher is empirically addressed by using longitudinal field experiences. We take a narrative approach and tell three stories from the field. In analysing the stories we propose a transition of the role of the engaged researcher, from that of a translator of general theory and contextual practices to one of a literary change agent. The literary change agent inspires practitioners by means as concepts, metaphors and storytelling. We suggest that the narrative approach can bring a new and critical flavour to the previously under-explored area of the role of the researcher in action research.
