A significant aspect of primary hepatic carcinoma in man is the high positive correlation of hepatocellular carcinoma with infection with hepatitis B virus (HBV)1. Analysis of the relationship between HBV infection and oncogenesis is difficult because natural infection with HBV is limited to man and experimental infection has been achieved only in chimpanzees and gibbons. Furthermore, because HBV has not been successfully propagated in cell culture, basic study of virus-cell interaction of the aetiological agent of one of the most widespread infections of man has been impossible. Recently, however, a cell line (PLC/PRF/5) derived from a human hepatoma biopsy was described which produces the HRV surface antigen (HBsAg) and so provides a tool for the experimental investigation of HBV in viro. We now report the derivation and characterisation of two additional cell lines primary liver carcinomas. In contrast to the PLC/PRF/5 cell line, these cell lines retain the capacity to synthesise many human plasma proteins, including both albumin and alpha-fetoprotein (AFP). One of these lines also produces BHsAg. We also present evidence that HBsAg synthesis and secretion in this cell line are correlated with the growth state of the culture. This finding is in contrast to the continuous HBsAg production found in the PLC/PRF/5 cell line.  相似文献

Post-meiotic segregation of alleles, which is seen, for example, in the 5:3 distribution of alleles in the products of a single meiosis in fungi, has been thought to be due to the non-repair of heteroduplex regions formed during genetic recombination. In current models of genetic recombination, heteroduplex DNA is formed either as the primary intermediate generated by two interacting non-sister chromatids or as a short region flanking a double-stranded gap. The frequency of post-meiotic segregation differs for different alleles, and this is presumed to reflect the varying efficiencies with which different types of mismatches in the heteroduplex are repaired. To gain some insight into this process, we have now determined the nucleotide sequences of various yeast alleles with different post-meiotic segregation frequencies and compared the mismatches predicted to occur in heteroduplexes of these alleles with wild-type DNA with those repaired with varying efficiency in bacterial systems. A striking correlation is observed, with the mismatches predicted for high post-meiotic segregation frequency alleles being similar to mismatches repaired with low efficiency in bacteria. These results support the view that postmeiotic segregation frequency reflects heteroduplex repair efficiency and the contention that meiotic gene conversion is the result of the successful repair of heteroduplex mismatches.  相似文献

Patchwork homology observed between divergent members of polymorphic multigene families is thought to reflect evolution by short-tract gene conversion (nonreciprocal recombination), although this mechanism cannot usually be confirmed in higher organisms. In contrast to meiotic conversions observed in laboratory yeast strains, apparent conversions between polymorphic sequences, such as the class I loci of the major histocompatibility complex (MHC), are short and do not seem to be associated with reciprocal recombination (crossover, exchanges). We have now integrated two nonallelic murine class I genes into yeast to characterize their meiotic recombination. We found no crossovers between the MHC genes, but short-tract 'microconversions' of 1-215 base-pairs were observed in about 6% of all meioses. Strikingly, one of these events was accompanied by a single base-pair mutation. These results underscore both the importance of meiotic gene conversion and sequence heterology in determining conversion patterns between divergent genes.  相似文献

Formalizing the separability condition in Bell's theorem   总被引:1,自引:0,他引:1  
The nonseparability of physical systems is often invoked in philosophical analyses of what has come to be known as Bell's theorem. Until recently, the formalization of the notion of separability was assumed to be unproblematic, equivalent to that of outcome independence (Jarrett incompleteness). Although this equivalence has been called into question, an alternative has not yet been specified with sufficient precision, leading to confusion as to what kinds of models should be considered separable. I identify four plausible candidates for the proper formalization of the separability condition, understood in terms of part–whole determination, and then discuss the relative merits of each. I show that three of these are, in fact, equivalent to outcome independence under most conditions, and that one is not, raising the possibility of a new decomposition of Bell locality. I also question whether part–whole determination should be considered sufficient for separability.  相似文献

Accelerated evolution of a false-truffle from a mushroom ancestor   总被引:5,自引:0,他引:5

THE false-truffles (Hymenogastrales) are a group of basidomycetous fungi that produce underground truffle-like basidiocarps. They are generally believed to be independently derived from several mushroom lineages, but extensive morphological divergence often obscures recognition of these phylogenetic connections. Comparisons of mitochondrial DNA now demonstrate a surprisingly close relationship between species of false-truffles in the genus Rhizopogon (Hymenogastraceae) and the mushroom genus Suillus (Boletaceae). The striking morphological differences separating all Suillus species from Rhizopogon imply an acceleration in the rate of morphological change relative to molecular change during the evolution of these false-truffles from their mushroom ancestors. This acceleration can best be explained by rapid morphological divergence resulting from selective pressures which may have acted on a small number of developmental genes.  相似文献

Two different models have been proposed to explain the relative frequencies of the non-mendelian allelic segregations which are detected by tetrad analysis after meiosis in fungi. The first model maintains that 6:2 type tetrads result from correction of heteroduplexes containing mismatched sites and 5:3 type tetrads result from failure to correct mismatched sites. The second model suggests that 6:2 segregations result from the filling-in of double-strand gaps using information obtained from both strands of a homologous duplex. In this model 5:3 type tetrads result if the allele is included in the heteroduplex regions flanking the gap and the resulting mismatched nucleotides are not corrected. We have studied the correction of heteroduplex plasmid DNA in pms1 mutant strains of Saccharomyces cerevisiae, which are known to exhibit higher frequencies of 5:3 type tetrads and lower frequencies of 6:2 tetrads than wild-type strains. Our results suggest that the pms1 mutation causes a defect in mismatch correction, supporting the hypothesis that meiotic gene conversion in wild-type yeast cells often results from the correction of heteroduplex DNA.  相似文献

CD24 is a glycosyl-phosphatidylinositol-anchored membrane protein that is frequently over-expressed in a variety of human carcinomas and is correlated with poor prognosis. In cancer cell lines, changes of CD24 expression can alter several cellular properties in vitro and tumor growth in vivo. However, little is known about how CD24 mediates these effects. Here we have analyzed the functional consequences of CD24 knock-down or over-expression in human cancer cell lines. Depletion of CD24 reduced cell proliferation and adhesion, enhanced apoptosis, and regulated the expression of various genes some of which were identified as STAT3 target genes. Loss of CD24 reduced STAT3 and FAK phosphorylation. Diminished STAT3 activity was confirmed by specific reporter assays. We found that reduced STAT3 activity after CD24 knock-down was accompanied by altered Src phosphorylation. Silencing of Src, similar to CD24, targeted the expression of prototype STAT3-regulated genes. Likewise, the over-expression of CD24 augmented Src-Y416 phosphorylation, the recruitment of Src into lipid rafts and the expression of STAT3-dependent target genes. An antibody to CD24 was effective in reducing tumor growth of A549 lung cancer and BxPC3 pancreatic cancer xenografts in mice. Antibody treatment affected the level of Src-phosphorylation in the tumor and altered the expression of STAT3 target genes. Our results provide evidence that CD24 regulates STAT3 and FAK activity and suggest an important role of Src in this process. Finally, the targeting of CD24 by antibodies could represent a novel route for tumor therapy.  相似文献

滑动型颈椎前路固定板在骨科临床上得到逐步应用,以取代限制型固定板并用来承担椎体上载荷及防止植骨下沉。然而,目前骨科临床对两种前路固定板的综合生物力学性能评价很少。研究使用18具猪颈椎骨模拟椎体切除手术,使用MTSMini858实验台模拟人体颈椎生理运动,采用三维运动分析系统、微型压力传感器分别测定椎体运动及椎间压力,实验评价滑动型与限制型前路固定板的生物力学性能。实验数据表明,滑动型固定板在两节段椎体次全切除术中的综合生物力学特性优于限制型前路固定板。  相似文献
