This final reply responds to Honohan’s invitation to articulate the Arendtian tone of the key-note paper. It spells out the philosophical intuition that the political life of citizens, at least potentially, is capable of making visible what makes human life worthwhile and fully meaningful, and the philosophical curiosity to see whether traces of this deep political awareness can be retrieved in dialogues with volunteers. In response to Dekker’s critical doubts, this final reply clarifies the central stakes of Claes’s paper. The core argument was not to show that the biographical model of meaningfulness is the prevailing approach of meaning in/of volunteering, but to assess the potentials and limits of the model’s interpretive power. Moreover, the paper argues for an alternative, existential model of meaningfulness. This approach refers to deep experiences of meaning that emerge from the practice of volunteering and that shift into powerful political experiences of hope, and a lived sense of equality.  相似文献

Recent genome-wide association studies have described many loci implicated in type 2 diabetes (T2D) pathophysiology and β-cell dysfunction but have contributed little to the understanding of the genetic basis of insulin resistance. We hypothesized that genes implicated in insulin resistance pathways might be uncovered by accounting for differences in body mass index (BMI) and potential interactions between BMI and genetic variants. We applied a joint meta-analysis approach to test associations with fasting insulin and glucose on a genome-wide scale. We present six previously unknown loci associated with fasting insulin at P < 5 × 10(-8) in combined discovery and follow-up analyses of 52 studies comprising up to 96,496 non-diabetic individuals. Risk variants were associated with higher triglyceride and lower high-density lipoprotein (HDL) cholesterol levels, suggesting a role for these loci in insulin resistance pathways. The discovery of these loci will aid further characterization of the role of insulin resistance in T2D pathophysiology.  相似文献

This article argues, firstly, that voluntary civic practices are not doomed to fall prey to a Big Society rhetoric and a cynical politics of cuts in social spending. It all depends on how these civic practices are promoted and what kind of civic discourse is communicated through the channels of social media and public opinion. This introductory article starts by describing the genesis of this special issue and the interconnection of its topics. The editors offer a variety of reading entries into the key-note articles and responses. The article reconstructs the research interests underpinning the idea of integrating meaningfulness, volunteers and citizenship. It highlights the explicit interdisciplinary design of the special issue, and shows how the key-note authors, and their respondents, weave connections between meaningfulness, volunteering and citizenship. And, finally, the editors bring the background understandings of the key-note papers to the foreground, and reconstruct a non-intentional meta-level discussion on two fundamental concepts and their interplay: self and world.  相似文献

This paper starts from qualitative research on volunteering and citizenship, with a special focus on volunteering within a setting of restorative justice and mediation. In a first stage, the author reconstructs two models of meaningfulness as hermeneutical lenses to better understand how volunteers see their engagement and experiences as a source of meaningfulness. The paper argues that a biographical model of meaningfulness (Wolf’s theoretical framework) is in need of a complementary approach to meaningfulness (an existential model of meaningfulness), which focuses on transformational experiences with a strong existential depth. In a second stage, the paper dialogues with the experiences and narratives of the volunteers, and shows how their understanding of citizenship informs, and transforms their ideas of meaningfulness. Both the biographical and the existential approach to meaningfulness appear to be deeply influenced by the volunteers’ civic imaginaries.  相似文献

Neuroleukin (NLK) is a protein of relative molecular mass (Mr) 56,000 (56K) secreted by denervated rat muscle and found in large amounts in muscle, brain, heart and kidneys. The protein is a neurotrophic factor for spinal and sensory neurons and a lymphokine product of lectin-stimulated T-cells. It also induces immunoglobulin secretion by human mononuclear cells. Molecular clones of NLK have been expressed in monkey COS cells and the product was shown to have the same biological and biochemical properties as the extracted protein. NLK is abundant in muscle, brain and kidney, but is active at concentrations of 10(-9) to 10(-11) M, similar to those for other polypeptide factors. We have cloned the gene for pig muscle phosphohexose isomerase (PHI) (EC 5.3.1.9) which catalyses the conversion of glucose-6-phosphate to fructose-6-phosphate, an obligatory step in glycolysis, and determined its amino-acid sequence. Surprisingly, it is 90% homologous to the sequence of mouse neuroleukin.  相似文献

Blood vessels and nerves are complex, branched structures that share a high degree of anatomical similarity. Guidance of vessels and nerves has to be exquisitely regulated to ensure proper wiring of both systems. Several regulators of axon guidance have been identified and some of these are also expressed in endothelial cells; however, the extent to which their guidance functions are conserved in the vascular system is still incompletely understood. We show here that the repulsive netrin receptor UNC5B is expressed by endothelial tip cells of the vascular system. Disruption of the Unc5b gene in mice, or of Unc5b or netrin-1a in zebrafish, leads to aberrant extension of endothelial tip cell filopodia, excessive vessel branching and abnormal navigation. Netrin-1 causes endothelial filopodial retraction, but only when UNC5B is present. Thus, UNC5B functions as a repulsive netrin receptor in endothelial cells controlling morphogenesis of the vascular system.  相似文献
