In multivariate discrimination of several normal populations, the optimal classification procedure is based on quadratic discriminant functions. We compare expected error rates of the quadratic classification procedure if the covariance matrices are estimated under the following four models: (i) arbitrary covariance matrices, (ii) common principal components, (iii) proportional covariance matrices, and (iv) identical covariance matrices. Using Monte Carlo simulation to estimate expected error rates, we study the performance of the four discrimination procedures for five different parameter setups corresponding to standard situations that have been used in the literature. The procedures are examined for sample sizes ranging from 10 to 60, and for two to four groups. Our results quantify the extent to which a parsimonious method reduces error rates, and demonstrate that choosing a simple method of discrimination is often beneficial even if the underlying model assumptions are wrong.The authors wish to thank the editor and three referees for their helpful comments on the first draft of this article. Cell stress such as hypoxia elicits adaptive responses, also on the level of mitochondria, and in part is mediated by the hypoxia-inducible factor (HIF) 1α. Adaptation of mitochondria towards acute hypoxic conditions is reasonably well understood, while regulatory mechanisms, especially of respiratory chain assembly factors, under chronic hypoxia remains elusive. One of these assembly factors is transmembrane protein 126B (TMEM126B). This protein is part of the mitochondrial complex I assembly machinery. We identified changes in complex I abundance under chronic hypoxia, in association with impaired substrate-specific mitochondrial respiration. Complexome profiling of isolated mitochondria of the human leukemia monocytic cell line THP-1 revealed HIF-1α-dependent deficits in complex I assembly and mitochondrial complex I assembly complex (MCIA) abundance. Of all mitochondrial MCIA members, we proved a selective HIF-1-dependent decrease of TMEM126B under chronic hypoxia. Mechanistically, HIF-1α induces the E3-ubiquitin ligase F-box/WD repeat-containing protein 1A (β-TrCP1), which in turn facilitates the proteolytic degradation of TMEM126B. Attenuating a functional complex I assembly appears critical for cellular adaptation towards chronic hypoxia and is linked to destruction of the mitochondrial assembly factor TMEM126B.  相似文献

Membrane proteins are key elements in cell physiology and drug targeting, but getting a high-resolution structure by crystallographic means is still enormously challenging. Novel strategies are in big demand to facilitate the structure determination process that will ultimately hasten the day when sequence information alone can provide a three-dimensional model. Cell-free or in vitro expression enables rapid access to large quantities of high-quality membrane proteins suitable for an array of applications. Despite its impressive efficiency, to date only two membrane proteins produced by the in vitro approach have yielded crystal structures. Here, we have analysed synergies of cell-free expression and crystallisation in lipid mesophases for generating an X-ray structure of the integral membrane enzyme diacylglycerol kinase to 2.28-Å resolution. The quality of cellular and cell-free-expressed kinase samples has been evaluated systematically by comparing (1) spectroscopic properties, (2) purity and oligomer formation, (3) lipid content and (4) functionality. DgkA is the first membrane enzyme crystallised based on cell-free expression. The study provides a basic standard for the crystallisation of cell-free-expressed membrane proteins and the methods detailed here should prove generally useful and contribute to accelerating the pace at which membrane protein structures are solved.  相似文献

Serum uric acid concentrations are correlated with gout and clinical entities such as cardiovascular disease and diabetes. In the genome-wide association study KORA (Kooperative Gesundheitsforschung in der Region Augsburg) F3 500K (n = 1,644), the most significant SNPs associated with uric acid concentrations mapped within introns 4 and 6 of SLC2A9, a gene encoding a putative hexose transporter (effects: -0.23 to -0.36 mg/dl per copy of the minor allele). We replicated these findings in three independent samples from Germany (KORA S4 and SHIP (Study of Health in Pomerania)) and Austria (SAPHIR; Salzburg Atherosclerosis Prevention Program in Subjects at High Individual Risk), with P values ranging from 1.2 x 10(-8) to 1.0 x 10(-32). Analysis of whole blood RNA expression profiles from a KORA F3 500K subgroup (n = 117) showed a significant association between the SLC2A9 isoform 2 and urate concentrations. The SLC2A9 genotypes also showed significant association with self-reported gout. The proportion of the variance of serum uric acid concentrations explained by genotypes was about 1.2% in men and 6% in women, and the percentage accounted for by expression levels was 3.5% in men and 15% in women.  相似文献

The bend in the Hawaiian-Emperor seamount chain is a prominent feature usually attributed to a change in Pacific plate motion approximately 47 Myr ago. However, global plate motion reconstructions fail to predict the bend. Here we show how the geometry of the Hawaiian-Emperor chain and other hotspot tracks can be explained when we combine global plate motions with intraplate deformation and movement of hotspot plumes through distortion by global mantle flow. Global mantle flow models predict a southward motion of the Hawaiian hotspot. This, in combination with a plate motion reconstruction connecting Pacific and African plates through Antarctica, predicts the Hawaiian track correctly since the date of the bend, but predicts the chain to be too far west before it. But if a reconstruction through Australia and Lord Howe rise is used instead, the track is predicted correctly back to 65 Myr ago, including the bend. The difference between the two predictions indicates the effect of intraplate deformation not yet recognized or else not recorded on the ocean floor. The remaining misfit before 65 Myr ago can be attributed to additional intraplate deformation of similar magnitude.  相似文献

There is concern that species loss may adversely affect ecosystem functioning and stability. But although there is evidence that biodiversity loss can lead to reductions in biomass production, there is no direct evidence that biodiversity loss affects ecosystem resistance (ability to withstand perturbation) or resilience (recovery from perturbation). Yet theory, laboratory experiments and indirect experimental evidence strongly suggest that diversity and stability are related. Here we report results from a field experiment with factorially crossed perturbation and diversity manipulations. We simulated drought perturbation on constructed grassland ecosystems containing 1, 2, 4, 8 or 32 plant species. Under unperturbed conditions, the species-poor systems achieved lower biomass production than the species-rich systems. However, the species-poor systems were more resistant to perturbation than the species-rich systems. The species-poor systems also showed a larger initial resilience following perturbation, although the original relationship between diversity and productivity was fully restored after 1year. Our results confirm that biodiversity increases biomass production, but they also point to the fact that such diversity--production associations may lead to an inverse relationship between biodiversity and the stability of ecosystem functioning.  相似文献
