Detecting myocardial ischemia with 2-D CVIB imaging method--an in vivo animal experiment study

A 2-D cyclic variation of integrated backscatter (CVIB) imaging method was established for detecting myocardial ischemia. To demonstrate the feasibility and validity of this method, animal experiments were conducted. Acute myocardial ischemia was induced by occluding left anterior descending coronary artery in 10 anesthetized open-chest dogs. While scanning the normal hearts and the ischemic hearts with a B scanner, digital radiofrequency data were acquired by a real-time acquisition system in synchronism. The offline analysis to the radio-frequency signal with the 2-D CVIB imaging method was performed to verify the consistency between the imaging result and the design of the experiment. In addition, 4 dogs in experiment were treated with the heart pacemaker in order to investigate the influence of changing in heart rate on the detection of ischemic myocardium with the proposed method. The experimental result showed that the 2-D CVIB imaging method succeeded in detecting the ischemic myocardium and is a new non-invasive way for the cardiologists to both quantitatively and visually evaluate the contractile performance of the myocardium.     

Icariin suppresses bone resorption activity of rabbit osteoclasts in vitro

The effect of icariin on the bone resorption activity of rabbit osteoclasts is assessed in vitro. Osteoclasts were isolated from Japanese white rabbits and cultured on plates with a sterilized bone slice in each well. After treatment with icariin at various concentrations, the bone resorption activity of osteoclasts was evaluated by examining pit areas, superoxide anion (·O2-) generation, size and number of actin rings and intracellular calcium concentration [Ca2 ]i. As revealed by these data, icariin elicited continuous decline of [Ca2 ]i, making actin ring constricted and ·O2- generation decreased. These events resulted in smaller and fewer pits which indicate suppressed bone resorption activity of rabbit osteoclasts by icariin.

Corrosion and electrochemical behavior of as-cast Mg-5Y-7Gd-1Nd-0.5Zr magnesium alloys in 5% NaCl aqueous solution

The corrosion mechanism of the as-cast Mg-5Y-7Gd-1Nd-0.5Zr magnesium alloy immersed in 5% NaCl aqueous solution was investigated by electrochemical test and SEM. The electrochemical results indicate that the corrosion resistance of Mg-5Y-7Gd-1Nd-0.5Zr alloy in 5% NaCl aqueous solution gradually deteriorates with immersion time increasing from 2 h, 24 h, 60 h, to 108 h, which is consistent with th e SEM observation of corrosion morphologies. The Cl^-anion leads to the initiation and development of the corrosion pits, and Mg-5Y-7Gd-1Nd-0.5Zr alloy exhibits filiform type of attack under significant anodic control of magnesium solution reaction. The corrosion potential becomes noble with increasing immersion time.     

Vanadyl ions binding to GroEL （HSP60） and inducing its depolymerization

Several vanadium compounds have been known for the hypoglycemic and anticancer effects. However, the mechanisms of the pharmacological and toxicological effects were not clear. In this work, we in- vestigated the potential targets of vanadium in mitochondria. Vanadyl ions were found to bind to mi- tochondria from rat liver with a stoichiometry of 244±58 nmol/mg protein and an apparent dissocia- tion constant (Kd) of (2.0±0.8)×10·16 mol/L. Using size exclusion chromatography, a vanadium-binding protein was isolated and identified to be the 60-kDa heat shock protein (HSP60) by mass spectrometry analysis and immunoassays. Additionally, binding of vanadyl ions was found to result in depolymeri- zation of homo-oligomeric HSP60 (GroEL). HSP60 is an indispensable molecular chaperone and in- volved in many kinds of pathogenesis of inflammatory and autoimmune diseases, e.g. type 1 diabetes. Our results suggested that HSP60 could be a novel important target involved in the biological and/or toxicological effects of vanadium compounds.

Permeation of vanadium（Ⅲ, Ⅳ, Ⅴ）-dipicolinate complexes across MDCK cell monolayer and comparison with Caco-2 cells

The permeation and eytotoxicity of three insulin-mimetic vanadium（Ⅲ, Ⅳ, Ⅴ）-dipicolinate complexes were studied using the MDCK cell monolayer in comparison with the Caeo-2 cells. On MDCK cell monolayer, the apparent permeation coefficients （Papp） were estimated to be （7.5±1.0）×10^-6, （1.0±0.2）×10^-6, （1.7±0.4）× 10^-6cm/s for V（Ⅴ）, V（Ⅵ）, and V（Ⅲ）-dipie complexes, respectively. The permeability of V（Ⅴ）-dipie complexes is much better than the others, which is in agreement with its better hypoglycemie effect in animal tests. On Caeo-2 cell monolayer, Papp were found to be in the range of 1-3×10^-6 ends and not to be affected by excessive amounts of dipieolinate ligand. By contrast, the permeability in the AP→BL direction across the MDCK monolayer increased greatly in the presence of free ligands, suggesting existence of active transport mechanism of vanadium complex anions on the MDCK cells. The eytotoxieity of the three complexes was found similar and the IC50 were measured in the range of 0.6-0.9 mmol/L for MDCK cells and 1.6--2 mmol/L for Caco-2 cells. The cytotoxicity of three vanadium complexes was conceivably in consistence with their permeability, suggesting that the toxicity, permeation and cellular metabolism of vanadium complexes are closely related.
