Multiple sclerosis (MS) is a complex chronic neurologic disease with a suspected autoimmune pathogenesis. Although there is evidence that the development of MS is determined by both environmental influences and genes, these factors are largely undefined, except for major histocompatibility (MHC) genes. Linkage analyses and association studies have shown that susceptibility to MS is associated with genes in the human histocompatibility leukocyte antigens (HLA) class II region, but the contribution of these genes to MS disease development less compared with their contribution to disorders such as insulin-dependent diabetes mellitus. Due to the strong linkage disequilibrium in the MHC class II region, it has not been possible to determine which gene(s) is responsible for the genetic predisposition. In transgenic mice, we have expressed three human components involved in T-cell recognition of an MS-relevant autoantigen presented by the HLA-DR2 molecule: DRA*0101/DRB1*1501 (HLA-DR2), an MHC class II candidate MS susceptibility genes found in individuals of European descent; a T-cell receptor (TCR) from an MS-patient-derived T-cell clone specific for the HLA-DR2 bound immunodominant myelin basic protein (MBP) 4102 peptide; and the human CD4 coreceptor. The amino acid sequence of the MBP 84-102 peptide is the same in both human and mouse MBP. Following administration of the MBP peptide, together with adjuvant and pertussis toxin, transgenic mice developed focal CNS inflammation and demyelination that led to clinical manifestations and disease courses resembling those seen in MS. Spontaneous disease was observed in 4% of mice. When DR2 and TCR double-transgenic mice were backcrossed twice to Rag2 (for recombination-activating gene 2)-deficient mice, the incidence of spontaneous disease increased, demonstrating that T cells specific for the HLA-DR2 bound MBP peptide are sufficient and necessary for development of disease. Our study provides evidence that HLA-DR2 can mediate both induced and spontaneous disease resembling MS by presenting an MBP self-peptide to T cells.  相似文献

It has long been known that there are two classes of gamma-ray bursts (GRBs), mainly distinguished by their durations. The breakthrough in our understanding of long-duration GRBs (those lasting more than approximately 2 s), which ultimately linked them with energetic type Ic supernovae, came from the discovery of their long-lived X-ray and optical 'afterglows', when precise and rapid localizations of the sources could finally be obtained. X-ray localizations have recently become available for short (duration <2 s) GRBs, which have evaded optical detection for more than 30 years. Here we report the first discovery of transient optical emission (R-band magnitude approximately 23) associated with a short burst: GRB 050709. The optical afterglow was localized with subarcsecond accuracy, and lies in the outskirts of a blue dwarf galaxy. The optical and X-ray afterglow properties 34 h after the GRB are reminiscent of the afterglows of long GRBs, which are attributable to synchrotron emission from ultrarelativistic ejecta. We did not, however, detect a supernova, as found in most nearby long GRB afterglows, which suggests a different origin for the short GRBs.  相似文献

Gamma-ray bursts (GRBs) come in two classes: long (> 2 s), soft-spectrum bursts and short, hard events. Most progress has been made on understanding the long GRBs, which are typically observed at high redshift (z approximately 1) and found in subluminous star-forming host galaxies. They are likely to be produced in core-collapse explosions of massive stars. In contrast, no short GRB had been accurately (< 10') and rapidly (minutes) located. Here we report the detection of the X-ray afterglow from--and the localization of--the short burst GRB 050509B. Its position on the sky is near a luminous, non-star-forming elliptical galaxy at a redshift of 0.225, which is the location one would expect if the origin of this GRB is through the merger of neutron-star or black-hole binaries. The X-ray afterglow was weak and faded below the detection limit within a few hours; no optical afterglow was detected to stringent limits, explaining the past difficulty in localizing short GRBs.  相似文献

It is now accepted that long-duration gamma-ray bursts (GRBs) are produced during the collapse of a massive star. The standard 'collapsar' model predicts that a broad-lined and luminous type Ic core-collapse supernova accompanies every long-duration GRB. This association has been confirmed in observations of several nearby GRBs. Here we report that GRB 060505 (ref. 10) and GRB 060614 (ref. 11) were not accompanied by supernova emission down to limits hundreds of times fainter than the archetypal supernova SN 1998bw that accompanied GRB 980425, and fainter than any type Ic supernova ever observed. Multi-band observations of the early afterglows, as well as spectroscopy of the host galaxies, exclude the possibility of significant dust obscuration and show that the bursts originated in actively star-forming regions. The absence of a supernova to such deep limits is qualitatively different from all previous nearby long-duration GRBs and suggests a new phenomenological type of massive stellar death.  相似文献

为解决深海X70管线钢在实际焊接中粗晶区(CGHAZ)的脆化问题,在不同热循环工艺下对X70管线钢进行了热模拟研究。采用Gleeble-3800热模拟机模拟X70管线钢CGHAZ,研究CGHAZ在10~60 kJ/cm不同热输入(HI)条件下组织和韧性的变化规律,并通过光学显微镜(OM)、扫描电镜(SEM)和夏比冲击试验等手段表征CGHAZ的组织和韧性。结果表明,不同热输入下试验钢的组织主要由粒状贝氏体(GB)、贝氏体铁素体(BF)和马-奥组元(M-A组元)组成;当HI不断增大时,BF比例减少,GB比例增加,M-A组元粗化,冲击吸收能先升高再降低;当HI为20 kJ/cm时,BF和GB可获得优异组合,断口为韧性断裂,冲击吸收能达到173.8 J;当HI大于20 kJ/cm时,断口解离断裂,冲击吸收能下降明显,最低为18.8 J。因此,较低的热输入可提高CGHAZ的韧性,使X70管线钢具有高强度、高韧性和良好的焊接性。研究结果可为优化焊接工艺提供理论依据。  相似文献

针对高超声速飞行器在临近空间巡航时出现的通信"黑障"问题,根据RAM C提供的飞行试验数据,建立一维等离子体鞘套模型,通过数值计算分析了等离子体与太赫兹波的相互作用机理,并从等离子体厚度、等离子体电子密度、等离子体碰撞频率和太赫兹波入射角等条件得到了太赫兹波在等离子体鞘套中的传输特性曲线。仿真结果表明:把太赫兹波段作为临近空间平台通信,有利于解决"黑障"问题,其中在大气窗口0.22THz处的衰减均在30dB以下。此论证结果可为临近空间平台设计的高超声速飞行器选用通信频段时提供参考。  相似文献

The theoretical framework of cosmology is mainly defined by gravity, of which general relativity is the current model. Recent tests of general relativity within the Lambda Cold Dark Matter (ΛCDM) model have found a concordance between predictions and the observations of the growth rate and clustering of the cosmic web. General relativity has not hitherto been tested on cosmological scales independently of the assumptions of the ΛCDM model. Here we report an observation of the gravitational redshift of light coming from galaxies in clusters at the 99 per cent confidence level, based on archival data. Our measurement agrees with the predictions of general relativity and its modification created to explain cosmic acceleration without the need for dark energy (the f(R) theory), but is inconsistent with alternative models designed to avoid the presence of dark matter.  相似文献

考察了加入新的活性污泥使解体好氧颗粒污泥完成修复的可行性.解体好氧颗粒污泥对新加入的活性污泥进行吸附,在各种选择压力特别是水力剪切力作用下和原有颗粒污泥形成一个有机整体.大约3周时间,解体颗粒污泥被完全修复.扫描电镜观察发现,被修复后的颗粒污泥呈现非常规则的结构,微生物相十分致密.在解体颗粒污泥逐渐被修复的过程中,颗粒平均粒径仅从最初的2.8mm增至2.9mm,说明活性污泥在颗粒污泥上的附着主要发生在颗粒的空穴.而颗粒污泥的沉降性能和强度都得到了极大的改善,颗粒沉降速率和完整性系数分别由最初的72m/h和56.8%提高到110m/h和65.8%.新加入的活性污泥除了部分用于修复解体颗粒污泥,其余在选择压力等的作用下形成了新的好氧颗粒污泥.  相似文献

When massive stars exhaust their fuel, they collapse and often produce the extraordinarily bright explosions known as core-collapse supernovae. On occasion, this stellar collapse also powers an even more brilliant relativistic explosion known as a long-duration gamma-ray burst. One would then expect that these long gamma-ray bursts and core-collapse supernovae should be found in similar galactic environments. Here we show that this expectation is wrong. We find that the gamma-ray bursts are far more concentrated in the very brightest regions of their host galaxies than are the core-collapse supernovae. Furthermore, the host galaxies of the long gamma-ray bursts are significantly fainter and more irregular than the hosts of the core-collapse supernovae. Together these results suggest that long-duration gamma-ray bursts are associated with the most extremely massive stars and may be restricted to galaxies of limited chemical evolution. Our results directly imply that long gamma-ray bursts are relatively rare in galaxies such as our own Milky Way.  相似文献
