It has been unclear whether amorphous glassy water heated to around 140-150 K remains glassy until it crystallizes or whether instead it turns into a supercooled and very viscous liquid. Yue and Angell compare the behaviour of glassy water under these conditions to that of hyperquenched inorganic glasses, and claim that water stays glassy as it heats up to its crystallization point; they also find a 'hidden' glass-to-liquid transition at about 169 K. Here we use differential scanning calorimetry (DSC) heating to show that hyperquenched water deposited at 140 K behaves as an ultraviscous liquid, the limiting structure of which depends on the cooling rate--as predicted by theoretical analysis of the liquid-to-glass transition. Our findings are consistent with a glass-to-liquid transition-onset temperature (T(g)) in the region of 136 K (refs 3,4), and they indicate that measurements of the liquid's properties may clarify the anomalous properties of supercooled water.  相似文献

Summary DNA sequence interspersion was investigated in pheasant and pelican nuclear DNA. As is typical for birds, these genomes are organized in a long period pattern. Altogether, 5 bird species with genome sizes between 1.6 and 1.9 pg DNA are compared with regard to the extent of repetitive and single copy sequence interspersion. Pathogenic bacteria often use effector molecules to increase virulence. In most cases, the mode of action of effectors remains unknown. Strains of Pseudomonas syringae pv. syringae (Pss) secrete syringolin A (SylA), a product of a mixed non-ribosomal peptide/polyketide synthetase, in planta. Here we identify SylA as a virulence factor because a SylA-negative mutant in Pss strain B728a obtained by gene disruption was markedly less virulent on its host, Phaseolus vulgaris (bean). We show that SylA irreversibly inhibits all three catalytic activities of eukaryotic proteasomes, thus adding proteasome inhibition to the repertoire of modes of action of virulence factors. The crystal structure of the yeast proteasome in complex with SylA revealed a novel mechanism of covalent binding to the catalytic subunits. Thus, SylA defines a new class of proteasome inhibitors that includes glidobactin A (GlbA), a structurally related compound from an unknown species of the order Burkholderiales, for which we demonstrate a similar proteasome inhibition mechanism. As proteasome inhibitors are a promising class of anti-tumour agents, the discovery of a novel family of inhibitory natural products, which we refer to as syrbactins, may also have implications for the development of anti-cancer drugs. Homologues of SylA and GlbA synthetase genes are found in some other pathogenic bacteria, including the human pathogen Burkholderia pseudomallei, the causative agent of melioidosis. It is thus possible that these bacteria are capable of producing proteasome inhibitors of the syrbactin class.  相似文献

Hepatocellular carcinoma is generally refractory to clinical treatment. Here, we report that inactivation of the MYC oncogene is sufficient to induce sustained regression of invasive liver cancers. MYC inactivation resulted en masse in tumour cells differentiating into hepatocytes and biliary cells forming bile duct structures, and this was associated with rapid loss of expression of the tumour marker alpha-fetoprotein, the increase in expression of liver cell markers cytokeratin 8 and carcinoembryonic antigen, and in some cells the liver stem cell marker cytokeratin 19. Using in vivo bioluminescence imaging we found that many of these tumour cells remained dormant as long as MYC remain inactivated; however, MYC reactivation immediately restored their neoplastic features. Using array comparative genomic hybridization we confirmed that these dormant liver cells and the restored tumour retained the identical molecular signature and hence were clonally derived from the tumour cells. Our results show how oncogene inactivation may reverse tumorigenesis in the most clinically difficult cancers. Oncogene inactivation uncovers the pluripotent capacity of tumours to differentiate into normal cellular lineages and tissue structures, while retaining their latent potential to become cancerous, and hence existing in a state of tumour dormancy.  相似文献

In most populations of mountain goats ( Oreamnos americanus ), mineral lick use is an essential part of the ecology of the species. In many areas, the distribution and use of licks in the landscape is poorly known, rendering planning for resource development difficult. We examined lick use by 28 GPS radio-collared mountain goats in 2 study areas in southeastern British Columbia during 2004–2005. Viewing collar-location movements on digital orthophotos, we assumed goat use of 6 previously known and 10 suspected mineral licks. Field visits verified that 9 of the 10 suspected sites were mineral licks. Thirteen of the 15 licks used by collared goats were within forests with commercial harvesting potential. All but 3 of the licks were ≤600 m from the closest logging block, and 5 licks were <100 m away. Number of annual visits to licks by individual goats ranged from 0 to 9. Goats often moved considerable distances (up to 17.3 km) to visit licks. Most visits by males occurred between early May and late June (median 9 June), and most visits by females occurred between early June and mid-July (median 21 June). Mean time spent at licks on each visit was 1.5 days for females and 1.6 days for males. Most of the licks were characterized by numerous cavities dug under trees (which we term “lick trees”). Using GPS collars, we were able to collect data on lower-elevation mineral licks not previously known to researchers.  相似文献

Summary Crosses betweenMicroseris pygmaea (10 pappus parts per fruit) andM. bigelovii (5 pappus parts) have revealed 10-determining genes that additively determine the average number of pappus parts in the hybrids. Summary Peripheral blood human T (TL) and B (BL) lymphocytes were separated by continuous Ficoll-Hypaque (FH) gradients. BL were found at the 1050 density interfase (70.5% EAC rosettes) with no TL (0% EAC rosettes); while at the 1068 density interfase there was an enrichment of TL (68% E rosettes) with very few BL (8.5% EAC rosettes).  相似文献

Summary Highly differentiated ZZ/ZW sex chromosomes and an exceptionally low genome, size were found in the karyotypes ofPyxicephalus adspersus (Anura, Ranidae). The W-chromosome is considerably smaller than the Z-chromosome and consists to a very great proportion of constitutive heterochromatin. The DNA content of this species and the chromosome length have the lowest values determined in the Ranidae to date.This study was supported by a grant from the Deutsche Forschungsgemeinschaft. The polarized architecture of epithelial cells depends on the highly stereotypic distribution of cellular junctions and other membrane-associated protein complexes. In epithelial cells of the Drosophila embryo, three distinct domains subdivide the lateral plasma membrane. The most apical one comprises the subapical complex (SAC). It is followed by the zonula adherens (ZA) and, further basally, by the septate junction. A core component of the SAC is the transmembrane protein Crumbs, the cytoplasmic domain of which recruits the PDZ-protein Discs Lost into the complex. Cells lacking crumbs or the functionally related gene stardust fail to organize a continuous ZA and to maintain cell polarity. Here we show that stardust provides an essential component of the SAC. Stardust proteins colocalize with Crumbs and bind to the carboxy-terminal amino acids of its cytoplasmic tail. We introduce two different Stardust proteins here: one MAGUK protein, characterized by a PDZ domain, an SH3 domain and a guanylate kinase domain; and a second isoform comprising only the guanylate kinase domain. The Stardust proteins represent versatile candidates as structural and possibly regulatory constituents of the SAC, a crucial element in the control of epithelial cell polarity.  相似文献
