Summary Contrary to earlier findings in rats, cerebral lateral ventricular infusions of 1×10–3 M or 2×10–3 M solutions of phloridzin at a rate of 2.5 l/min for 90 min had no significant stimulating effects on food intake and weight gain in hens and cocks. These different responses to intraventricular phloridzin might reflect a difference of sensitivity to the inhibitory action of phloridzin on glucose transport in cerebral cells or certain peculiarities of mechanisms controlling food intake in chickens. 相似文献

The receptor (CXCR4) for the stromal-derived factor-1 (SDF1) and the urokinase-receptor (uPAR) are up-regulated in various tumors. We show that CXCR4-transfected cells migrate toward SDF1 on collagen (CG) and do not on vitronectin (VN). Co-expression of cell-surface uPAR, which is a VN receptor, impairs SDF1-induced migration on CG and allows migration on VN. Blocking fMLP receptors (fMLP-R), alpha-v integrins or the uPAR region capable to interact with fMLP-Rs, impairs migration of uPAR/CXCR4-transfected cells on VN and restores their migration on CG. uPAR co-expression also reduces the adherence of CXCR4-expressing cells to various components of the extracellular matrix (ECM) and influences the partitioning of beta1 and alpha-v integrins to membrane lipid-rafts, affecting ECM-dependent signaling. uPAR interference in CXCR4 activity has been confirmed in cells from prostate carcinoma. Our results demonstrate that uPAR expression regulates the adhesive and migratory ability of CXCR4-expressing cells through a mechanism involving fMLP receptors and alpha-v integrins. 相似文献

Recognized as a “disease modifier”, physical activity (PA) is increasingly viewed as a more holistic, cost-saving method for prevention, treatment and management of human disease conditions. The traditional view that PA engages the monoaminergic and endorphinergic systems has been challenged by the discovery of the endocannabinoid system (ECS), composed of endogenous lipids, their target receptors, and metabolic enzymes. Indeed, direct and indirect evidence suggests that the ECS might mediate some of the PA-triggered effects throughout the body. Moreover, it is now emerging that PA itself is able to modulate ECS in different ways. Against this background, in the present review we shall discuss evidence of the cross-talk between PA and the ECS, ranging from brain to peripheral districts and highlighting how ECS must be tightly regulated during PA, in order to maintain its beneficial effects on cognition, mood, and nociception, while avoiding impaired energy metabolism, oxidative stress, and inflammatory processes. 相似文献

Calorie restriction (CR) extends lifespan in a wide spectrum of organisms and is the only regimen known to lengthen the lifespan of mammals. We established a model of CR in budding yeast Saccharomyces cerevisiae. In this system, lifespan can be extended by limiting glucose or by reducing the activity of the glucose-sensing cyclic-AMP-dependent kinase (PKA). Lifespan extension in a mutant with reduced PKA activity requires Sir2 and NAD (nicotinamide adenine dinucleotide). In this study we explore how CR activates Sir2 to extend lifespan. Here we show that the shunting of carbon metabolism toward the mitochondrial tricarboxylic acid cycle and the concomitant increase in respiration play a central part in this process. We discuss how this metabolic strategy may apply to CR in animals. 相似文献

异常黑胆质成熟剂是维吾尔医复方制剂,由甘草、红枣、破布木、牛舌草、铁线蕨、地锦草、小茴香、薰衣草、蜜蜂花和刺糖10味药组成,用于治疗由异常黑胆质所导致的肿瘤、糖尿病和心血管疾病等。研究了异常黑胆质成熟剂中各单味药(除刺糖外)对HL-60细胞增殖的抑制作用。HL-60细胞暴露于各单味药两种不同提取物(二氯甲烷和甲醇),于37℃、CO2体积分数为5%的恒温孵箱中培养后24、48、72h,用台盼蓝染色法检测细胞存活率。结果表明,各单味药的二氯甲烷提取物除小茴香外均显著降低HL-60癌细胞的存活率(P<0.05)。在甲醇提取物中,只有甘草和地锦草两味药的甲醇提取物可显著降低HL-60癌细胞的存活率(P<0.05)。与各单味药甲醇提取物相比,其二氯甲烷提取物均显著降低HL-60癌细胞的存活率(P<0.05)。结果显示,破布木、地锦草、铁线蕨、甘草、红枣、薰衣草、牛舌草和蜜蜂花8味药的二氯甲烷提取物和甘草、地锦草两味药的甲醇提取物均能抑制HL-60癌细胞的体外增殖。各单味药二氯甲烷提取物对HL-60细胞增殖的抑制作用均明显强于其相应的甲醇提取物。 相似文献

Polydnavirus-encoded IκB-like proteins are similar to insect and mammalian IκB, and an immunosuppressive function in the host
cells has been inferred to these proteins. Here we show that the expression of one of these IκB-like viral genes, the TnBVank1, in the Drosophila germline affects the localization of gurken, bicoid, and oskar mRNAs whose gene products are relevant for proper embryonic patterning. The altered localization of these mRNAs is suggestive
of general defects in the intracellular, microtubule-based, trafficking routes. Analysis of microtubule motor proteins components
such as the dynein heavy chain and the kinesin heavy chain revealed defects in the polarized microtubule network. Interestingly,
the TnBVANK1 viral protein is uniformly distributed over the entire oocyte cortex, and appears to be anchored to the microtubule

Platelets are stored at 22°C, since incubation at 37°C results in loss of viability. Nonetheless, in our body (37°C), platelets
survive for 8–10 days. This discrepancy has been explained in terms of deprivation of viability factors or accumulation of
apoptotic factors during storage. We report that the endocannabinoid anandamide (AEA) may be one of the agents allowing platelet
survival. In fact, at 37°C, human platelets enhance the expression of pro-apoptotic proteins (caspases, Bax, Bak) and decrease
the expression of Bcl-xL, thus changing the Bcl-xL/Bak ratio, a key platelet biological clock. AEA or its non-hydrolyzable
analogue, methanandamide, extend platelet life span, without reversing the changes in Bcl-xL/Bak ratio induced by heat stress.
Instead, AEA binding to type-1 cannabinoid receptor activates Akt, which regulates, through phosphorylation of Bad, the interactions
among different Bcl-2 family members. These findings could have implications for platelet collection and, potentially, for

At central excitatory synapses, N-methyl-D-aspartate (NMDA) receptors, which have a high affinity for glutamate, produce a slowly rising synaptic current in response to a single transmitter pulse and an additional current after a second, closely timed stimulus. Here we show, by examining the kinetics of transmitter binding and channel gating in single-channel currents from recombinant NR1/NR2A receptors, that the synaptic response to trains of impulses is determined by the molecular reaction mechanism of the receptor. The rate constants estimated for the activation reaction predict that, after binding neurotransmitter, receptors hesitate for approximately 4 ms in a closed high-affinity conformation before they either proceed towards opening or release neurotransmitter, with about equal probabilities. Because only about half of the initially fully occupied receptors become active, repetitive stimulation elicits currents with distinct waveforms depending on pulse frequency. This high-affinity/low-efficiency activation mechanism might serve as a link between stimulation frequency and the directionality of the ensuing synaptic plasticity. 相似文献
