The endocannabinoid system and endocannabinoid receptor-driven modulation of glutamate release were studied in rat brain cortex astroglial gliosomes. These preparations contained the endocannabinoids N-arachidonoylethanolamine (anandamide) and 2-arachidonoylglycerol, as well their major biosynthetic (N-acyl-phosphatidylethanolamines-hydrolyzing-phospholipase D and diacylglycerol-lipase) and catabolic (fatty acid amide-hydrolase and monoacylglycerol-lipase) enzymes. Gliosomes expressed type-1 (CB1R), type-2 (CB2R) cannabinoid, and type-1 vanilloid (TRPV1) receptors, as ascertained by Western blotting and confocal microscopy. Methanandamide, a stable analogue of anandamide acting as CB1R, CB2R, and TRPV1 agonist, stimulated or inhibited the depolarization-evoked gliosomal [3H]d-aspartate release, at lower and higher concentrations, respectively. Experiments with ACEA (arachidonyl-2′-chloroethylamide), JWH133 ((6aR,10aR)-3-(1,1-dimethylbutyl)-6a,7,10,10a-tetrahydro-6,6,9-trimethyl-6H-dibenzo[b,d]-pyran) and capsaicin, selective agonists at CB1R, CB2R and TRPV1, respectively, demonstrated that potentiation of [3H]d-aspartate release was due to CB1R while inhibition to CB2R and TRPV1 engagement. These findings were confirmed by using selective receptor antagonists. Furthermore, CB1R activation caused increase of intracellular IP3 and Ca2+ concentration, suggesting an involvement of phospholipase C.  相似文献

In the early Universe, while galaxies were still forming, black holes as massive as a billion solar masses powered quasars. Supermassive black holes are found at the centres of most galaxies today, where their masses are related to the velocity dispersions of stars in their host galaxies and hence to the mass of the central bulge of the galaxy. This suggests a link between the growth of the black holes and their host galaxies, which has indeed been assumed for a number of years. But the origin of the observed relation between black hole mass and stellar velocity dispersion, and its connection with the evolution of galaxies, have remained unclear. Here we report simulations that simultaneously follow star formation and the growth of black holes during galaxy-galaxy collisions. We find that, in addition to generating a burst of star formation, a merger leads to strong inflows that feed gas to the supermassive black hole and thereby power the quasar. The energy released by the quasar expels enough gas to quench both star formation and further black hole growth. This determines the lifetime of the quasar phase (approaching 100 million years) and explains the relationship between the black hole mass and the stellar velocity dispersion.  相似文献

Large-scale applications of high-transition-temperature (high-T(c)) superconductors, such as their use in superconducting cables, are impeded by the fact that polycrystalline materials (the only practical option) support significantly lower current densities than single crystals. The superconducting critical current density (J(c)) across a grain boundary drops exponentially if the misorientation angle exceeds 2 degrees -7 degrees. Grain texturing reduces the average misorientation angle, but problems persist. Adding impurities (such as Ca in YBa2Cu3O7-delta; YBCO) leads to increased J(c) (refs 9, 10), which is generally attributed to excess holes introduced by Ca2+ substituting for Y3+ (ref. 11). However, a comprehensive physical model for the role of grain boundaries and Ca doping has remained elusive. Here we report calculations, imaging and spectroscopy at the atomic scale that demonstrate that in poly-crystalline YBCO, highly strained grain-boundary regions contain excess O vacancies, which reduce the local hole concentration. The Ca impurities indeed substitute for Y, but in grain-boundary regions under compression and tension they also replace Ba and Cu, relieving strain and suppressing O-vacancy formation. Our results demonstrate that the ionic radii are more important than their electronic valences for enhancing J(c).  相似文献

In On Local Motion in the Two New Sciences, Galileo distinguishes between ‘time’ and ‘quanto time’ to justify why a variation in speed has the same properties as an interval of time. In this essay, I trace the occurrences of the word quanto to define its role and specific meaning. The analysis shows that quanto is essential to Galileo’s mathematical study of infinitesimal quantities and that it is technically defined. In the light of this interpretation of the word quanto, Evangelista Torricelli’s theory of indivisibles can be regarded as a natural development of Galileo’s insights about infinitesimal magnitudes, transformed into a geometrical method for calculating the area of unlimited plane figures.  相似文献

Summary In 21 rabbits, the average concentration of sodium and potassium in the aqueous humor was found to be respectively 149±4.3 mEq/l and 5.34±mEq/l.Eleven of these animals were injected with Diamox (100 mg/kg of body weight); a significant drop of potassium concentration in the aqueous humor was observed three hours later, as compared with the ten control animals. No change in sodium concentration occurred.Our findings are considered in correlation with the fall of intraocular pressure produced by the carbonic anhydrase inhibitor, and the role of this enzyme is discussed.  相似文献
