Experimental autoimmune encephalomyelitis (EAE) is a prototype autoimmune disease mediated by type 1 helper T (TH1) cells and under the control of regulatory cells. Here we report that a synthetic glycolipid ligand for CD1d-restricted natural killer T (NKT) cells expressing the semi-invariant T-cell receptor (Valpha14+) is preventive against EAE. The ligand is an analogue of alpha-galactosylceramide (alpha-GC), a prototype NKT cell ligand, with a truncated sphingosine chain. alpha-GC causes NKT cells to produce both interferon (IFN)-gamma and interleukin (IL)-4 (refs 4, 5). However, this new ligand can induce a predominant production of IL-4 by the NKT cells. A single injection of this glycolipid, but not of alpha-GC, consistently induced TH2 bias of autoimmune T cells by causing NKT cells to produce IL-4, leading to suppression of EAE. The lack of polymorphism of CD1d and cross-reactive response of mouse and human NKT cells to the same ligand indicates that targeting NKT cells with this ligand may be an attractive means for intervening in human autoimmune diseases such as multiple sclerosis.  相似文献

Cyclic electron flow around photosystem I is essential for photosynthesis   总被引:4,自引:0,他引:4  
Photosynthesis provides at least two routes through which light energy can be used to generate a proton gradient across the thylakoid membrane of chloroplasts, which is subsequently used to synthesize ATP. In the first route, electrons released from water in photosystem II (PSII) are eventually transferred to NADP+ by way of photosystem I (PSI). This linear electron flow is driven by two photochemical reactions that function in series. The cytochrome b6f complex mediates electron transport between the two photosystems and generates the proton gradient (DeltapH). In the second route, driven solely by PSI, electrons can be recycled from either reduced ferredoxin or NADPH to plastoquinone, and subsequently to the cytochrome b6f complex. Such cyclic flow generates DeltapH and thus ATP without the accumulation of reduced species. Whereas linear flow from water to NADP+ is commonly used to explain the function of the light-dependent reactions of photosynthesis, the role of cyclic flow is less clear. In higher plants cyclic flow consists of two partially redundant pathways. Here we have constructed mutants in Arabidopsis thaliana in which both PSI cyclic pathways are impaired, and present evidence that cyclic flow is essential for efficient photosynthesis.  相似文献

Summary Upon injection of -phenylethylamine (PEA) into the subconjunctival space of rabbit, intraocular pressure increased. This paper examines the history of two related problems concerning earthquakes, and the way in which a theoretical advance was involved in their resolution. The first problem is the development of a physical, as opposed to empirical, scale for measuring the size of earthquakes. The second problem is that of understanding what happens at the source of an earthquake. There was a controversy about what the proper model for the seismic source mechanism is, which was finally resolved through advances in the theory of elastic dislocations. These two problems are linked, because the development of a physically-based magnitude scale requires an understanding of what goes on at the seismic source. I will show how the theoretical advances allowed seismologists to re-frame the questions they were trying to answer, so that the data they gathered could be brought to bear on the problem of seismic sources in new ways.  相似文献

By exome sequencing, we found de novo SMARCB1 mutations in two of five individuals with typical Coffin-Siris syndrome (CSS), a rare autosomal dominant anomaly syndrome. As SMARCB1 encodes a subunit of the SWItch/Sucrose NonFermenting (SWI/SNF) complex, we screened 15 other genes encoding subunits of this complex in 23 individuals with CSS. Twenty affected individuals (87%) each had a germline mutation in one of six SWI/SNF subunit genes, including SMARCB1, SMARCA4, SMARCA2, SMARCE1, ARID1A and ARID1B.  相似文献

The interactions of metal ions with nucleic acids, nucleosides, and nucleo-bases have been extensively investigated .We have reported that thymine-thymine (T-T) and cytosine-cytosine (C-C) miss base pairs in DNA duplexes highly selectively capture Hgn ion and AgI ion, which result in formations of metal-mediated base pairs, T-Hg^Ⅱ -T and C-Ag^Ⅰ -C, in duplexes. The phenomenon is expected to be useful for a variety of studies such as synthesis of nano-wires containing metal ions, developing metal-ion sensing methods, etc. Here, we report novel oligodeoxyribonucleotide (ODN)-based sensors that detect Hgn ions and Ag^Ⅰ ions in aqueous solutions.  相似文献
