DNA fingerprinting transforms the art of cell authentication.

The increasing diversity of new cell cultures is seriously stretching the capabilities of traditional methods of identification. DNA fingerprinting is set to play an important role in increasing confidence in the authenticity of cultures in research and industry.     

Exercise conditioning decreases downstream movement of pond-reared razorback suckers released into a stream environment

Despite augmentation stocking efforts, wild populations of razorback suckers ( Xyrauchen texanus ) continue to decline. Endangered razorback suckers are commonly raised in off-channel ponds until maturity (approximately 300 mm TL) and then stocked into the Colorado River or its tributaries. After fish are stocked, they commonly move large distances downstream. We conducted an experiment to determine if downstream dispersal could be reduced through exercise conditioning. Two groups of razorback suckers, exercised and nonexercised, were released into Fossil Creek, Arizona. Prior to release, a subsample from each treatment group was tested in a laboratory flow chamber. Razorback suckers that had been exercise conditioned were able to maintain a position in the flow chamber 2 times longer and at velocities 25 cm · s –1 higher than nonexercised fish. Although the intended method of field data collection via passive-integrated-transponder (PIT) antennas and a remote communication station failed because river otters ( Lontra canadensis ) preyed upon the released razorback suckers, implanted PIT tags were retained in otter scat. Recovered PIT tags enabled distributional analysis, which indicated that exercised razorback suckers did not move as far downstream from the point of release as nonexercised razorbacks. Exercise conditioning may increase overall fitness of pond-reared razorback suckers, and, consequently, increase the effectiveness of augmentation stocking. PDGF signalling controls age-dependent proliferation in pancreatic β-cells

Determining the signalling pathways that direct tissue expansion is a principal goal of regenerative biology. Vigorous pancreatic β-cell replication in juvenile mice and humans declines with age, and elucidating the basis for this decay may reveal strategies for inducing β-cell expansion, a long-sought goal for diabetes therapy. Here we show that platelet-derived growth factor receptor (Pdgfr) signalling controls age-dependent β-cell proliferation in mouse and human pancreatic islets. With age, declining β-cell Pdgfr levels were accompanied by reductions in β-cell enhancer of zeste homologue 2 (Ezh2) levels and β-cell replication. Conditional inactivation of the Pdgfra gene in β-cells accelerated these changes, preventing mouse neonatal β-cell expansion and adult β-cell regeneration. Targeted human PDGFR-α activation in mouse β-cells stimulated Erk1/2 phosphorylation, leading to Ezh2-dependent expansion of adult β-cells. Adult human islets lack PDGF signalling competence, but exposure of juvenile human islets to PDGF-AA stimulated β-cell proliferation. The discovery of a conserved pathway controlling age-dependent β-cell proliferation indicates new strategies for β-cell expansion.     

Common variants on 1p36 and 1q42 are associated with cutaneous basal cell carcinoma but not with melanoma or pigmentation traits

To search for new sequence variants that confer risk of cutaneous basal cell carcinoma (BCC), we conducted a genome-wide SNP association study of 930 Icelanders with BCC and 33,117 controls. After analyzing 304,083 SNPs, we observed signals from loci at 1p36 and 1q42, and replicated these associations in additional sample sets from Iceland and Eastern Europe. Overall, the most significant signals were from rs7538876 on 1p36 (OR = 1.28, P = 4.4 x 10(-12)) and rs801114 on 1q42 (OR = 1.28, P = 5.9 x 10(-12)). The 1p36 locus contains the candidate genes PADI4, PADI6, RCC2 and ARHGEF10L, and the gene nearest to the 1q42 locus is the ras-homolog RHOU. Neither locus was associated with fair pigmentation traits that are known risk factors for BCC, and no risk was observed for melanoma. Approximately 1.6% of individuals of European ancestry are homozygous for both variants, and their estimated risk of BCC is 2.68 times that of noncarriers.
