Poly[d(G-C)] serves as the prototype for the right-to-left (B to Z) transition in he helical sense of DNA, both in solution and inthe crystal form. However, the question remains as to which other synthetic and natural DNAs have the potential to adopt left-handed conformations. One logical candidate is the canonical alternating purine-pyrimidine sequence d(A-C)n.d(G-T)n which seems to be widely disseminated in eukaryotic genomes. Our approach to this question is based on the enzymatic synthesis of poly[d(A-C).d(G-U)] derivatives with systematic methyl and halogen substitutions in the C-5 position of the pyrimidines C and U. Such modifications in poly[d(G-C)] have previously been shown to potentiate the B to Z transition. Here we report a highly cooperative, reversible, salt- and temperature-dependent transition for poly[d(A-m5C).d(G-T)], a repeat of the d(A-m5C) sequence which may occur in natural DNA. Spectroscopic studies and the demonstrated ability to bind anti-Z-DNA antibodies suggest that the new helical conformation is left-handed and shares structural features with known Z-DNA. However, a novel property, not exhibited by poly[d(G-C)], is the profound temperature dependence of the conformational equilibrium. 相似文献

Intraperitoneal injection of zinc raised levels of a hepatic metallothionein-like species. Assuming that this species was metallothionein (MT) then levels were raised from approximately 20 micrograms/g to 300 micrograms/g in 7 days, and levels thereafter remained high for the next 4 weeks. The half-lives of the protein in liver and kidney from starved fish, measured using in vivo incorporation of 35S cysteine at 11 degrees C, were approximately 27 days and 32 days respectively. The following agents failed to stimulate synthesis of MT in plaice: stress (due to catching), endotoxin, dexamethasone, cortisol and turpentine. 相似文献

The neonatal Fc receptor (FcRn) transports maternal IgG across epithelial barriers, thereby providing the fetus or newborn with humoral immunity before its immune system is fully functional. In newborn rats, FcRn transfers IgG from milk to blood by apical-to-basolateral transcytosis across intestinal epithelial cells. The pH difference between the apical (pH 6.0-6.5) and basolateral (pH 7.4) sides of intestinal epithelial cells facilitates the efficient unidirectional transport of IgG, because FcRn binds IgG at pH 6.0-6.5 but not at pH 7 or more. As milk passes through the neonatal intestine, maternal IgG is removed by FcRn-expressing cells in the proximal small intestine (duodenum and jejunum); remaining proteins are absorbed and degraded by FcRn-negative cells in the distal small intestine (ileum). Here we use electron tomography to make jejunal transcytosis visible directly in space and time, developing new labelling and detection methods to map individual nanogold-labelled Fc within transport vesicles and simultaneously to characterize these vesicles by immunolabelling. Combining electron tomography with a non-perturbing endocytic label allowed us to conclusively identify receptor-bound ligands, resolve interconnecting vesicles, determine whether a vesicle was microtubule-associated, and accurately trace FcRn-mediated transport of IgG. Our results present a complex picture in which Fc moves through networks of entangled tubular and irregular vesicles, only some of which are microtubule-associated, as it migrates to the basolateral surface. New features of transcytosis are elucidated, including transport involving multivesicular body inner vesicles/tubules and exocytosis through clathrin-coated pits. Markers for early, late and recycling endosomes each labelled vesicles in different and overlapping morphological classes, revealing spatial complexity in endo-lysosomal trafficking. 相似文献

Summary The role of renal hydrodynamics on renal deposition of immune complexes was evaluated in acute serum sickness. At mid-ocean ridges with fast to intermediate spreading rates, the upper section of oceanic crust is composed of lavas overlying a sheeted dyke complex. These units are formed by dykes intruding into rocks overlying a magma chamber, with lavas erupting at the ocean floor. Seismic reflection data acquired over young oceanic crust commonly image a reflector known as 'layer 2A', which is typically interpreted as defining the geologic boundary between lavas and dykes. An alternative hypothesis is that the reflector is associated with an alteration boundary within the lava unit. Many studies have used mapped variability in layer 2A thickness to make inferences regarding the geology of the oceanic crust, including volcanic construction, dyke intrusion and faulting. However, there has been no link between the geologic and seismological structure of oceanic crust except at a few deep drill holes. Here we show that, although the layer 2A reflector is imaged near the top of the sheeted dyke complex at fast-spreading crust located adjacent to the Hess Deep rift, it is imaged significantly above the sheeted dykes section at intermediate-spreading crust located near the Blanco transform fault. Although the lavas and underlying transition zone thicknesses differ by about a factor of two, the shallow seismic structure is remarkably similar at the two locations. This implies that seismic layer 2A cannot be used reliably to map the boundary between lavas and dykes in young oceanic crust. Instead we argue that the seismic layer 2A reflector corresponds to an alteration boundary that can be located either within the lava section or near the top of the sheeted dyke complex of oceanic crust. 相似文献
