Membrane proteins classically are handled in aqueous solutions as complexes with detergents. The dissociating character of detergents, combined with the need to maintain an excess of them, frequently results in more or less rapid inactivation of the protein under study. Over the past few years, we have endeavored to develop a novel family of surfactants, dubbed amphipols (APs). APs are amphiphilic polymers that bind to the transmembrane surface of the protein in a noncovalent but, in the absence of a competing surfactant, quasi-irreversible manner. Membrane proteins complexed by APs are in their native state, stable, and they remain water-soluble in the absence of detergent or free APs. An update is presented of the current knowledge about these compounds and their demonstrated or putative uses in membrane biology. 相似文献

In this study, a proteomic approach that combines selective labelling of proteins containing reduced cysteine residues with
two-dimensional electrophoresis/mass spectrometry was used to evaluate the redox state of protein cysteines during chronological
ageing in Saccharomyces cerevisiae. The procedure was developed on the grounds that biotinconjugated iodoacetamide (BIAM) specifically reacts with reduced cysteine
residues. BIAM-labelled proteins can then be selectively isolated by streptavidin affinity capture. We compared cells grown
on 2% glucose in the exponential phase and during chronological ageing and we found that many proteins undergo cysteine oxidation.
The target proteins include enzymes involved in glucose metabolism. Both caloric restriction and growth on glycerol resulted
in a decrease in the oxidative modification. Furthermore, in these conditions a reduced production of ROS and a more negative
glutathione half cell redox potential were observed.

Summary A new kind of hermaphroditism, which is called inducible hermaphroditism, has been discovered in the otherwise gonochoristic populations ofOphryotrocha labronica of the Gulf of Naples and of the Lagune of Venice. Inducible hermaphrodites, which are fertile in both the male and female phases, represent 25.8% and 31.8% of the Naples and Venice strains respectively. Inducible hermaphrodites are obtained when pairs between adult females — or female phase hermaphrodites — and juveniles are formed. Crosses between inducible hermaphrodites, in different sex phases, pure male and female individuals give progenies whose sex ratios are clearly correlated with the sex genotypes of the parents. 相似文献
