We present an alternative approach to Multiple Correspondence Analysis (MCA) that is appropriate when the data consist of ordered categorical variables. MCA displays objects (individuals, units) and variables as individual points and sets of category points in a low-dimensional space. We propose a hybrid decomposition on the basis of the classical indicator super-matrix, using the singular value decomposition, and the bivariate moment decomposition by orthogonal polynomials. When compared to standard MCA, the hybrid decomposition will give the same representation of the categories of the variables, but additionally, we obtain a clear association interpretation among the categories in terms of linear, quadratic and higher order components. Moreover, the graphical display of the individual units will show an automatic clustering.  相似文献

Dysfunction of the exocrine pancreas is observed in diabetes, but links between concurrent exocrine and endocrine pancreatic disease and contributing genetic factors are poorly characterized. We studied two families with diabetes and exocrine pancreatic dysfunction by genetic, physiological and in vitro functional studies. A genome-wide screen in Family 1 linked diabetes to chromosome 9q34 (maximal lod score 5.07). Using fecal elastase deficiency as a marker of exocrine pancreatic dysfunction refined the critical chromosomal region to 1.16 Mb (maximal lod score 11.6). Here, we identified a single-base deletion in the variable number of tandem repeats (VNTR)-containing exon 11 of the carboxyl ester lipase (CEL) gene, a major component of pancreatic juice and responsible for the duodenal hydrolysis of cholesterol esters. Screening subjects with maturity-onset diabetes of the young identified Family 2, with another single-base deletion in CEL and a similar phenotype with beta-cell failure and pancreatic exocrine disease. The in vitro catalytic activities of wild-type and mutant CEL protein were comparable. The mutant enzyme was, however, less stable and secreted at a lower rate. Furthermore, we found some evidence for an association between common insertions in the CEL VNTR and exocrine dysfunction in a group of 182 unrelated subjects with diabetes (odds ratio 4.2 (1.6, 11.5)). Our findings link diabetes to the disrupted function of a lipase in the pancreatic acinar cells.  相似文献

SH-SY5Y neuroblastoma cells were cultured for up to three serial passages in the presence of the copper chelator triethylene tetramine (Trien). The copper-depleted neuroblastoma cell line obtained showed decreased activities of the copper enzymes Cu, Zn superoxide dismutase and cytochrome c oxidase with concomitant increases in reactive oxygen species. Mitochondrial antioxidants (Mn superoxide dismutase and Bcl-2) were up-regulated. Overexpression and activation of p53 were early responses, leading to an increase in p21. Eventually, copper-depleted cells detached from the monolayer and underwent apoptosis. Activation of up-stream caspase-9, but not caspase-8, suggested that apoptosis proceeds via a mitochondrial pathway, followed by caspase-3 activation. The addition of copper sulfate to the copper-depleted cells restored copper enzymes, normalized antioxidant levels and improved cell viability. We conclude that prolonged copper starvation in these replicating cells leads to mitochondrial damage and oxidative stress and ultimately, apoptosis.Received 24 April 2003; accepted 23 May 2003  相似文献

In this paper we present a way of conducting design of experiments by Multivariate Additive Partial Least-Squares Splines models, in short MAPLSS. In the framework of optimal experimental design based on small samples, in order to select the most informative MAPLSS model, we process an adaptive incremental selection of observations by a particular bootstrap procedure. Why MAPLSS models? Because they inherit the advantages of the PLS regression that permits to capture additively non-linear main effects and relevant interactions in the difficult framework of small samples. The effectiveness of this approach is illustrated on the reservoir simulator data used to forecast oil production.  相似文献

Summary Treatment of orchidectomized rats with LH, FSH or prolactin decreased the tubulin content of the medial basal hypothalamus (MBH), whereas FSH or prolactin augmented it in the adenohypophysis (AH). After castration, negative correlations existed between serum LH and total or polymerized MBH tubulin, whereas in the AH positive correlations were found.
