Summary In the dog and guinea-pig submandibular glands kallikrein seems to be present in the striated duct cells. Following sympathetic nerve and in vivo isoproterenol stimulation of the dog and guinea-pig submandibular gland respectively, there is a reduction of kallikrein concentration. Ultrastructurally this reduction corresponds to the decrease of straited duct secretory granules in both species. The type III receptor tyrosine kinase FLT3 is frequently mutated in acute myeloid leukemia. Oncogenic FLT3 mutants display constitutive activity leading to aberrant cell proliferation and survival. Phosphorylation on several critical tyrosine residues is known to be essential for FLT3 signaling. Among these tyrosine residues, Y842 is located in the so-called activation loop. The position of this tyrosine residue is well conserved in all receptor tyrosine kinases. It has been reported that phosphorylation of the activation loop tyrosine is critical for catalytic activity for some but not all receptor tyrosine kinases. The role of Y842 residue in FLT3 signaling has not yet been studied. In this report, we show that Y842 is not important for FLT3 activation or ubiquitination but plays a critical role in regulating signaling downstream of the receptor as well as controlling receptor stability. We found that mutation of Y842 in the FLT3-ITD oncogenic mutant background reduced cell viability and increased apoptosis. Furthermore, the introduction of the Y842 mutation in the FLT3-ITD background led to a dramatic reduction in in vitro colony forming capacity. Additionally, mice injected with cells expressing FLT3-ITD/Y842F displayed a significant delay in tumor formation, compared to FLT3-ITD expressing cells. Microarray analysis comparing gene expression regulated by FLT3-ITD versus FLT3-ITD/Y842F demonstrated that mutation of Y842 causes suppression of anti-apoptotic genes. Furthermore, we showed that cells expressing FLT3-ITD/Y842F display impaired activity of the RAS/ERK pathway due to reduced interaction between FLT3 and SHP2 leading to reduced SHP2 activation. Thus, we suggest that Y842 is critical for FLT3-mediated RAS/ERK signaling and cellular transformation.  相似文献

Chemotaxis is the directed motility by means of which microbes sense chemical cues and relocate towards more favorable environments. Methyl-accepting chemotaxis proteins (MCPs) are the most common receptors in bacteria and archaea. They are arranged as trimers of dimers that, in turn, form hexagonal arrays in the cytoplasmic membrane or in the cytoplasm. Several different classes of MCPs have been identified according to their ligand binding region and membrane topology. MCPs have been further classified based on the length and sequence conservation of their cytoplasmic domains. Clusters of membrane-embedded MCPs often localize to the poles of the cell, whereas cytoplasmic MCPs can be targeted to the poles or distributed throughout the cell body. MCPs play an important role in cell survival, pathogenesis, and biodegradation. Bacterial adaptation to diverse environmental conditions promotes diversity among the MCPs. This review summarizes structure, classification, and structure–activity relationship of the known MCP receptors, with a brief overview of the signal transduction mechanisms in bacteria and archaea.  相似文献

Past research indicates that forecasting is important in understanding price dynamics across assets. We explore the potentiality of multiscale forecasting in the crude oil market by employing a wavelet multiscale analysis on returns and volatilities of Brent and West Texas Intermediate crude oil indices between January 1, 2001, and May 1, 2015. The analysis is based on a shift-invariant discrete wavelet transform, augmented by an entropy-based methodology for determining the optimal timescale decomposition under different market regimes. The empirical results show that the five-step-ahead wavelet forecast that is based on volatilities outperforms the random walk forecast, relative to the wavelet forecast that is based on returns. Optimal wavelet causality forecasting for returns is suggested across all frequencies (i.e., daily–yearly), whereas for volatilities it is suggested only up to quarterly frequencies. These results may have important implications for market efficiency and predictability of prices on the crude oil markets.  相似文献

Summary Various procedures which reduce or deplete the kallikrein content of the cat's submandibular gland correspondingly reduce the number of apical granules in the striated duct cells. The kallikrein content is greatly reduced after chronic parasympathetic but not after sympathetic nerve section which suggests that the parasympathetic innervation is required for synthesis or storage of this enzyme.We wish to acknowledge the valuable assistance of Dr T. 利用大孔网状载体固定苯酚降解菌——布鲁氏杆菌(Brucella sp.)GXY-1菌株,并对固定化菌的制备条件及固定化菌降解苯酚的特性进行考察.结果表明,固定化菌株的最适温度为25~35℃,最适pH为7.0左右,摇床转速为150r/min,最大耐盐度为2%,比游离态菌株更能适应环境变化.固定化菌降解苯酚的速率明显高于游离菌,降解过程符合Andrews抑制模型,确定了模型参数:qmax=2.932h^-1,Ks=286.227mg/L,Ki=97.720mg/L.  相似文献
