Prokineticins are proteins that regulate diverse biological processes including gastrointestinal motility, angiogenesis, circadian rhythm, and innate immune response. Prokineticins bind two closed related G-protein coupled receptors (GPCRs), PKR1 and PKR2. In general, these receptors act as molecular switches to relay activation to heterotrimeric G-proteins and a growing body of evidence points to the fact that GPCRs exist as homo- or heterodimers. We show here by Western-blot analysis that PKR2 has a dimeric structure in neutrophils. By heterologous expression of PKR2 in Saccharomyces cerevisiae, we examined the mechanisms of intermolecular interaction of PKR2 dimerization. The potential involvement of three types of mechanisms was investigated: coiled-coil, disulfide bridges, and hydrophobic interactions between transmembrane domains. Characterization of differently deleted or site-directed PKR2 mutants suggests that dimerization proceeds through interactions between transmembrane domains. We demonstrate that co-expressing binding-deficient and signaling-deficient forms of PKR2 can re-establish receptor functionality, possibly through a domain-swapping mechanism.  相似文献

Cornelia de Lange syndrome is a multisystem developmental disorder characterized by facial dysmorphisms, upper limb abnormalities, growth delay and cognitive retardation. Mutations in the NIPBL gene, a component of the cohesin complex, account for approximately half of the affected individuals. We report here that mutations in SMC1L1 (also known as SMC1), which encodes a different subunit of the cohesin complex, are responsible for CdLS in three male members of an affected family and in one sporadic case.  相似文献

The motility and morphogenesis of endothelial cells is controlled by spatio-temporally regulated activation of integrin adhesion receptors, and integrin activation is stimulated by major determinants of vascular remodelling. In order for endothelial cells to be responsive to changes in activator gradients, the adhesiveness of these cells to the extracellular matrix must be dynamic, and negative regulators of integrins could be required. Here we show that during vascular development and experimental angiogenesis, endothelial cells generate autocrine chemorepulsive signals of class 3 semaphorins (SEMA3 proteins) that localize at nascent adhesive sites in spreading endothelial cells. Disrupting endogenous SEMA3 function in endothelial cells stimulates integrin-mediated adhesion and migration to extracellular matrices, whereas exogenous SEMA3 proteins antagonize integrin activation. Misexpression of dominant negative SEMA3 receptors in chick embryo endothelial cells locks integrins in an active conformation, and severely impairs vascular remodelling. Sema3a null mice show vascular defects as well. Thus during angiogenesis endothelial SEMA3 proteins endow the vascular system with the plasticity required for its reshaping by controlling integrin function.  相似文献

Summary Various restriction enzymes digest human highly repeated homogeneous DNA to discrete fragments, some of which are present in the male and absent in the female. Summary Satellite DNAs of theDrosophila willistoni group have been analyzed by Hae III endonuclease digestion. Differences were observed between the 2 speciesD. willistoni andD. paulistorum and also between semispecies ofD. paulistorum and, to a lesser extent, between subspecies ofD. willistoni. When clustering asymmetric proximity data, only the average amounts are often considered by assuming that the asymmetry is due to noise. But when the asymmetry is structural, as typically may happen for exchange flows, migration data or confusion data, this may strongly affect the search for the groups because the directions of the exchanges are ignored and not integrated in the clustering process. The clustering model proposed here relies on the decomposition of the asymmetric dissimilarity matrix into symmetric and skew-symmetric effects both decomposed in within and between cluster effects. The classification structures used here are generally based on two different partitions of the objects fitted to the symmetric and the skew-symmetric part of the data, respectively; the restricted case is also presented where the partition fits jointly both of them allowing for clusters of objects similar with respect to the average amounts and directions of the data. Parsimonious models are presented which allow for effective and simple graphical representations of the results.  相似文献

Summary Evidence of hybridization between 2 pairs of allopatricDolichopoda species is provided by electrophoretic analysis. Occurrence of hybrids was revealed by laboratory crosses and in nature both by occasional co-existence due to passive dispersal and by transplantation experiments.This research was supported by the National Research Council, Italy, grant No. 78.01441.04, and by funds of the Faculty of Sciences, University of Rome. The paper presents a methodology for classifying three-way dissimilarity data, which are reconstructed by a small number of consensus classifications of the objects each defined by a sum of two order constrained distance matrices, so as to identify both a partition and an indexed hierarchy. Specifically, the dissimilarity matrices are partitioned in homogeneous classes and, within each class, a partition and an indexed hierarchy are simultaneously fitted. The model proposed is mathematically formalized as a constrained mixed-integer quadratic problem to be fitted in the least-squares sense and an alternating least-squares algorithm is proposed which is computationally efficient. Two applications of the methodology are also described together with an extensive simulation to investigate the performance of the algorithm.  相似文献
