This report extends earlier work by Brailovsky on regression theory and methodology, giving particular emphasis to function approximation for incompletely specified models. The interest here is with situations where the form of the regression relation is not known in advance. We discuss several difficulties that arise in using local approximation and linear regression methods, and propose ways to overcome these problems. To aid the data analyst in developing a suitable model, an illustrative table is derived for determining the number of initial explanatory functions justifiable for a given prespecified confidence level. The general approach formulated here is illustrated with an application to medical data. Relevance to classification and possible extensions are discussed. 相似文献

Root hairs are cellular protuberances extending from the root surface into the soil; there they provide access to immobile inorganic ions such as phosphate, which are essential for growth. Their cylindrical shape results from a polarized mechanism of cell expansion called tip growth in which elongation is restricted to a small area at the surface of the hair-forming cell (trichoblast) tip. Here we identify proteins that spatially control the sites at which cell growth occurs by isolating Arabidopsis mutants (scn1) that develop ectopic sites of growth on trichoblasts. We cloned SCN1 and showed that SCN1 is a RhoGTPase GDP dissociation inhibitor (RhoGDI) that spatially restricts the sites of growth to a single point on the trichoblast. We showed previously that localized production of reactive oxygen species by RHD2/AtrbohC NADPH oxidase is required for hair growth; here we show that SCN1/AtrhoGDI1 is a component of the mechanism that focuses RHD2/AtrbohC-catalysed production of reactive oxygen species to hair tips during wild-type development. We propose that the spatial organization of growth in plant cells requires the local RhoGDI-regulated activation of the RHD2/AtrbohC NADPH oxidase. 相似文献

Iron is the most abundant transition-metal element in the mantle and therefore plays an important role in the geochemistry and geodynamics of the Earth's interior. Pressure-induced electronic spin transitions of iron occur in magnesiowüstite, silicate perovskite and post-perovskite. Here we have studied the spin states of iron in magnesiowüstite and the isolated effects of the electronic transitions on the elasticity of magnesiowüstite with in situ X-ray emission spectroscopy and X-ray diffraction to pressures of the lowermost mantle. An observed high-spin to low-spin transition of iron in magnesiowüstite results in an abnormal compressional behaviour between the high-spin and the low-spin states. The high-pressure, low-spin state exhibits a much higher bulk modulus and bulk sound velocity than the low-pressure, high-spin state; the bulk modulus jumps by approximately 35 percent and bulk sound velocity increases by approximately 15 percent across the transition in (Mg0.83,Fe0.17)O. Although no significant density change is observed across the electronic transition, the jump in the sound velocities and the bulk modulus across the transition provides an additional explanation for the seismic wave heterogeneity in the lowermost mantle. The transition also affects current interpretations of the geophysical and geochemical models using extrapolated or calculated thermal equation-of-state data without considering the effects of the electronic transition. 相似文献

Copernicus versucht, die Ptolemäischen Konstruktionen auf konzentrische Kreise und Epizykel zu reduzieren und aus ihnen ein wirklich einheitliches System zu schaffen. Bei der Umordnung, die er dazu vornimmt, bemüht er sich aber, so viel wie möglich vom Ptolemäischen System zu erhalten. Das punctum aequans ersetzt er bei den äußeren Planeten durch eine Näherungskonstruktion mit Hilfe eines Epizykels. Im Fall des Ptolemäischen Deferenten der Venus, der nach seinen Vorstellungen die Erdbahn wiedergibt, muß er einen dem punctum aequans annähernd gleichwertigen Epizykelmechanismus auf die Venusbahn verpflanzen.

Large igneous provinces (LIPs) are known for their rapid production of enormous volumes of magma (up to several million cubic kilometres in less than a million years), for marked thinning of the lithosphere, often ending with a continental break-up, and for their links to global environmental catastrophes. Despite the importance of LIPs, controversy surrounds even the basic idea that they form through melting in the heads of thermal mantle plumes. The Permo-Triassic Siberian Traps--the type example and the largest continental LIP--is located on thick cratonic lithosphere and was synchronous with the largest known mass-extinction event. However, there is no evidence of pre-magmatic uplift or of a large lithospheric stretching, as predicted above a plume head. Moreover, estimates of magmatic CO(2) degassing from the Siberian Traps are considered insufficient to trigger climatic crises, leading to the hypothesis that the release of thermogenic gases from the sediment pile caused the mass extinction. Here we present petrological evidence for a large amount (15?wt%) of dense recycled oceanic crust in the head of the plume and develop a thermomechanical model that predicts no pre-magmatic uplift and requires no lithospheric extension. The model implies extensive plume melting and heterogeneous erosion of the thick cratonic lithosphere over the course of a few hundred thousand years. The model suggests that massive degassing of CO(2) and HCl, mostly from the recycled crust in the plume head, could alone trigger a mass extinction and predicts it happening before the main volcanic phase, in agreement with stratigraphic and geochronological data for the Siberian Traps and other LIPs. 相似文献

The in vivo roles of meprin metalloproteases in pathophysiological conditions remain elusive. Substrates define protease roles. Therefore, to identify natural substrates for human meprin α and β we employed TAILS (terminal amine isotopic labeling of substrates), a proteomics approach that enriches for N-terminal peptides of proteins and cleavage fragments. Of the 151 new extracellular substrates we identified, it was notable that ADAM10 (a disintegrin and metalloprotease domain-containing protein 10)—the constitutive α-secretase—is activated by meprin β through cleavage of the propeptide. To validate this cleavage event, we expressed recombinant proADAM10 and after preincubation with meprin β, this resulted in significantly elevated ADAM10 activity. Cellular expression in murine primary fibroblasts confirmed activation. Other novel substrates including extracellular matrix proteins, growth factors and inhibitors were validated by western analyses and enzyme activity assays with Edman sequencing confirming the exact cleavage sites identified by TAILS. Cleavages in vivo were confirmed by comparing wild-type and meprin?/? mice. Our finding of cystatin C, elafin and fetuin-A as substrates and natural inhibitors for meprins reveal new mechanisms in the regulation of protease activity important for understanding pathophysiological processes. 相似文献

Titanium and its alloys are promising materials for hydrogen storage. However, hydrogen penetration accompanies the exploitation of hydrogen storage alloys. In particular, hydrogen penetration and accumulation in titanium alloys changes their mechanical properties. Therefore, the research works of such materials are mainly focused on improving the reversibility of hydrogen absorption-liberation processes, increasing the thermodynamic characteristics of the alloys, and augmenting their hydrogen storage capacity. In the process of hydrogenation-dehydrogenation, the formed defects both significantly reduce hydrogen storage capacity and can also be used to create effective traps for hydrogen. Therefore, the investigation of hydrogen interaction with structural defects in titanium and its alloys is very important. The present work, the hydrogen-induced formation of defects in the alloys of commercially pure titanium under temperature gas-phase hydrogenation(873 K) has studied by positron lifetime spectroscopy and Doppler broadening spectroscopy. Based on the evolution of positron annihilation parameters τ_f, τ_d, their corresponding intensities If, Idand relative changes of parameters S/S_0 and W/W_0, the peculiarities of hydrogen interaction with titanium lattice defects were investigated in a wide range of hydrogen concentrations from 0.8at% to 32.0at%. 相似文献

Stem-cell ageing is thought to contribute to altered tissue maintenance and repair. Older humans experience increased bone marrow failure and poorer haematologic tolerance of cytotoxic injury. Haematopoietic stem cells (HSCs) in older mice have decreased per-cell repopulating activity, self-renewal and homing abilities, myeloid skewing of differentiation, and increased apoptosis with stress. Here we report that the cyclin-dependent kinase inhibitor p16INK4a, the level of which was previously noted to increase in other cell types with age, accumulates and modulates specific age-associated HSC functions. Notably, in the absence of p16INK4a, HSC repopulating defects and apoptosis were mitigated, improving the stress tolerance of cells and the survival of animals in successive transplants, a stem-cell-autonomous tissue regeneration model. Inhibition of p16INK4a may ameliorate the physiological impact of ageing on stem cells and thereby improve injury repair in aged tissue. 相似文献
