The p53 protein integrates multiple upstream signals and functions as a tumor suppressor by activating distinct downstream genes. At the cellular level, p53 induces apoptosis, cell cycle arrest and senescence. A rare mutant form of p53 with the amino acid substitution R175P, found in human tumors, is completely defective in initiating apoptosis but still induces cell cycle arrest. To decipher the functional importance of these pathways in spontaneous tumorigenesis, we used homologous recombination to generate mice with mutant p53-R172P (the mouse equivalent of R175P in humans). Mice inheriting two copies of this mutation (Trp53(515C/515C)) escape the early onset of thymic lymphomas that characterize Trp53-null mice. At 7 months of age, 90% of Trp53-null mice had died, but 85% of Trp53(515C/515C) mice were alive and tumor-free, indicating that p53-dependent apoptosis was not required for suppression of early onset of spontaneous tumors. The lymphomas and sarcomas that eventually developed in Trp53(515C/515C) mice retained a diploid chromosome number, in sharp contrast to aneuploidy observed in tumors and cells from Trp53-null mice. The ability of mutant p53-R172P to induce a partial cell cycle arrest and retain chromosome stability are crucial for suppression of early onset tumorigenesis.  相似文献

Petroleum industries of Iran offer some of the best job opportunities within Iranian labor market. However, due to the attributes of the chemicals used in these industries, their staffs are exposed to various risk factors of several chronic diseases. Such exposures might lead to cancer incidence after a decade or two. The Ministry of Petroleum (MoP) runs an organization, namely Petroleum Industry Health Organization (PIHO), which is responsible for the health insurance of the personnel working in Petroleum industries. PIHO and Health, Safety & Environment (HSE) units play fundamental roles in providing the employees with health services during their professional life and retirement period. Yet, these organizations still do not have a system to specify occupational cancer control mechanisms at the MoP. The negative influences of cancer on patients suffering from it as well as its heavy costs of treatment have forced MoP to put measures of coping with this chronic disease in the first priority. This paper aims to define the structure and design a framework for MoP’s occupational cancer control problem as the first step for improving the described situation. Given the presence of various stakeholders and actors, and the wide range of complexities of this problematic situation, we have adopted Soft System Methodology (SSM). We have reached an agreement with the problem owners on a rich picture, CATWOE analysis, root definition and a conceptual model for the way this situation could improve within real world circumstances. The results were obtained through various sessions with practitioner from different departments of MoP, while the conflict of interests was common.  相似文献

Casing while drilling (CwD) technology is designed to reduce drilling time and expenses by improving the wellbore stability, fracture gradient, and formation damage while reducing the exposure time. However, for the purpose well control, the wellbore geometry and volumes differ from those obtained via a conventional drilling technique, thereby requiring a different approach. This study discusses well control principles for CwD operations. It presents a simplified method for evaluating the maximum kick tolerance and allowable well shut-in time for both conventional and CwD techniques using a mathematical model. Preliminary results revealed that the use of CwD leads to an annulus pressure loss three times higher than that observed in the conventional drilling. In addition, the kick tolerance is reduced by 50% and the maximum allowable well shut-in time is reduced by 65%, making an early kick detection system necessary.  相似文献

This paper proposes three fractional discrete chaotic systems based on the Rulkov, Chang,and Zeraoulia–Sprott rational maps. The dynamics of the proposed maps are investigated by means of phase plots and bifurcations diagrams. Adaptive stabilization schemes are proposed for each of the three maps and the convergence of the states is established by using the Lyapunov method. Furthermore, a combination synchronization scheme is proposed whereby a combination of the fractional Rulkov and Chang maps is synchronized to the fractional Zeraoulia-Sprott map. Numerical results are used to confirm the findings of the paper.  相似文献

This paper constructs a cyclic ?4-code with a parity-check matrix similar to that of Goethals code but in length 2m + 1, for all m ≥ 4. This code is a subcode of the lifted Zetterberg code for m even. Its minimum Lee weight is shown to be at least 10, in general, and exactly 12 in lengths 33, 65. The authors give an algebraic decoding algorithm which corrects five errors in these lengths for m = 5, 6 and four errors for m > 6.  相似文献
