Our understanding of the biochemical mechanisms that mediate chemoreception in insects has been greatly improved after the
discovery of olfactory and taste receptor proteins. However, the presence of soluble polypeptides in high concentration around
the dendrites of sensory neurons still poses unanswered questions. More than 2 decades after their discovery and despite the
wealth of structural information available, the physiological function of odorant-binding proteins is not well understood.
More recently, members of a second family of soluble polypeptides, the chemosensory proteins, were also discovered in the
lymph of chemosensilla. Here we review the structural properties of both classes of soluble proteins, their affinity to small
ligands, and their expression in the different parts of the insect body and subcellular localisation. Finally, we discuss
current ideas and models of the role of such proteins in insect chemoreception.

Ubiquitylated inclusion bodies (IBs) found in Huntington’s disease (HD) postulate an impaired ubiquitin-proteasome system.
However, this hypothesis remains controversial. In vitro-generated polyglutamine aggregates failed to inhibit purified proteasomes, while filamentous huntingtin aggregates isolated
from mice resulted in inhibition. However, similarly isolated IBs did not, thus suggesting that IB formation is protective
by sequestering smaller inhibitory aggregates. Accordingly, proteasome-activity assays in IB-containing mouse brain homogenates
did not show decreased activity. On the contrary, some of the endoproteolytic proteasome activities increased, probably due
to altered subunit composition. However, activity was found decreased in postmortem human HD tissue. Finally, evidence supporting
the hypothesis was found in HD cell models expressing fluorescent ubiquitin-proteasome system reporters but not in retina
of SCA-7 mice with similar reporters. In summary, it seems that mutant huntingtin, probably in intermediate aggregate forms,
has the potential to inhibit proteasome activity, but the global status of the system in HD brain tissue is not yet fully

The preparation of sufficient amounts of high-quality samples is still the major bottleneck for the characterization of membrane proteins by in vitro approaches. The hydrophobic nature, the requirement for complicated transport and modification pathways, and the often observed negative effects on membrane properties are intrinsic features of membrane proteins that frequently cause significant problems in overexpression studies. Establishing efficient protocols for the production of functionally folded membrane proteins is therefore a challenging task, and numerous specific characteristics have to be considered. In addition, a variety of expression systems have been developed, and choice of appropriate techniques could strongly depend on the desired target membrane proteins as well as on their intended applications. The production of membrane proteins is a highly dynamic field and new or modified approaches are frequently emerging. The review will give an overview of currently established processes for the production of functionally folded membrane proteins. 相似文献

Sperm competition is widespread and has played an important role in shaping male reproductive characters such as testis size and numbers of sperm produced, and this is reflected in the rapid evolution of many reproductive genes. Additionally, sperm competition has been implicated in the rapid evolution of seminal fluids. However, our understanding of the molecular basis of many traits thought to be important in sperm competition is rudimentary. Furthermore, links between sperm competition and a range of issues not directly related to reproduction are only just beginning to be explored. These include associations between sperm competition and selfish genes, immunity and diseases such as cancer.We briefly review these topics and suggest areas we consider worthy of additional research. 相似文献

Summary Substantial progress has been made in the last ten years in understanding the structural and functional organization of parasitic protozoa and helminths and the complex physiological relationships that exist between these organisms and their hosts. By employing the new powerful techniques of biochemistry, molecular biology and immunology the genomic organization in parasites, the molecular basis of parasite's variation in surface antigens and the biosynthesis, processing, transport and membrane anchoring of these and other surface proteins were extensively investigated. Significant advances have also been made in our knowledge of the specific and often peculiar strategies of intermediary metabolism, cell compartmentation, the role of oxygen for parasites and the mechanisms of antiparasitic drug action. Further major fields of interest are currently the complex processes which enables parasites to evade the host's immune defense system and other mechanisms which have resulted in the specific adaptations which enabled parasites to survive within their host environments. Various approaches in molecular and biochemical parasitology and in immunoparasitology have been proven to be of high potential for serodiagnosis, immunoprophylaxis and drug design.This paper is based on a review presented at a workshop on Molecular Parasitology, organized by the Swiss Society of Tropical Medecine and Parasitology at the University of Neuchâtel, March 1985. 相似文献

Numerous proteins in pro-and eukaryotes must cross cellular membranes in order to reach their site of function. Many of these proteins carry signal sequences that are removed by specific signal peptidases during, or shortly after, membrane transport. Signal peptidases have been identified in the rough endoplasmic reticulum, the matrix and inner membrane of mitochondria, the stroma and thylakoid membrane of chloroplasts, the bacterial plasma membrane and the thylakoid membrane of cyanobacteria. The composition of these peptidases varies between one and several subunits. No site-specific inhibitors are known for the majority of these enzymes. Accordingly, signal peptidases recognize structural motifs rather than linear amino acid sequences. Such motifs have become evident by employing extensive site-directed mutagenesis to investigate the anatomy of signal sequences. Analysis of the reaction specificities and the primary sequences of several signal peptidases suggests that the enzymes of the endoplasmic reticulum, the inner mitochondrial membrane and the thylakoid membrane of chloroplasts all have evolved from bacterial progenitors. 相似文献
