Babesia bovis: the effect of acute inflammation and isoantibody production in the detection of babesial antigens

Summary Immunoblots ofBabesia bovis antigen contain dominant antigens which react not only with antisera toB. bovis but with sera from naive calves recovering from an acute inflammatory reaction. It seems likely these antigens are from the host rather than the parasite.     

Cloning and expression of HIV-1 p24 gene in insect cells by using BAC-TO-BAC system

Recombinant transposing vector pFHIV24 was constructed by cloning the HIV-1 p24 gene into the Multiple cloning site (MCS) of the transposing vector pFastBac1 in the correct orientation with respect to the polyhedrin promoter. Recombinant bacmid bHIV24 was obtained by transposing a mini-att Tn7 element from the recombinant pFHIV24 to the mini-att Tn7 attachment site on the bacmid by Tn7 transposition functions provided by the helper plasmid. Minipreparation of recombinant bacmid DNA was transfected intoSpodoptera frugiperda (Sf9) cells to get the recombinant virus. Fresh insect Sf9 cells were infected with the recombinant virus containing p24 to express the target protein. The target protein expressed was analyzed on a 15% polyacrylamide gels and then used as antigen to check HIV-1 positive serum by ELISA. Our positive result shows that the expressed p24 protein could be used as standard antigen for HIV-1 diagnosis by ELISA and other reliable diagnostic methods of HIV-1 infection. What's new in the field of cancer vaccines?

The observation that in some cases tumors undergo spontaneous regression concomitantly with autoimmune manifestations has been interpreted as an indication of the involvement of the immune system in tumor rejection. This raised the conceptual possibility that the immune system could be used against the tumor. However, since tumor cells are poorly immunogenic by themselves, early attempts to develop immune-based approaches for cancer therapy saw the use of tumor cells transduced with genes coding for cytokines or costimulatory molecules to enhance in vivo immunity. The identification of cytotoxic T lymphocyte (CTL)-defined tumor associated antigens has allowed the development of new strategies for cancer immunotherapy. Novel adjuvants have been identified, and different modes of antigen delivery were devised which aim at inducing efficient CTL responses in patients. 检测前列腺特异抗原在前列腺疾病患者中临床价值的探讨

目的 探讨前列腺特异性抗原（PSA）浓度变化在前列腺疾病诊断中的应用价值。方法采用电化学发光免疫分析（ECL）法检测71例健康对照者、48例前列腺炎患者、111例良性前列腺增生（BPH）患者和59例前列腺癌（PCa）患者血清巾的PSA浓度，对检测结果进行统计分析：结果前列腺癌组患者血清PSA浓度明显高于其它各组（P〈0．001）；前列腺增生组部分病人血清PSA浓度升高，但局限在一定范围，一般不超过10．0mg/ml；前列腺炎组和健康对照组均在正常范嗣（〈4．0ng／ml）．结论PSA是筛查前列腺癌的灵敏的肿瘤标志物，对良、恶性前列腺疾病有一定的鉴别诊断意义。     

抗血吸虫循环抗原抗体制备的研究

采用不同的血吸虫抗原组分，经不同途径和方法免疫实验兔。结果显示微量免疫和Guo窝淋巴结内注射免疫的免疫效果好。Guo窝淋巴结内注射免疫，即使抗原被注入淋巴结附近仍可获得较好的免疫效果。抗甲基化载体蛋白可溶性成虫抗原抗体检测CSA的敏感度显著增高。甲基化载体成虫抗原加福氏完全佐剂和复合核苷酸钠佐剂的免疫成功率为8.50％，抗血清效价均在1：12800以上。     

大肠杆菌菌毛抗原K99基因的亚克隆及核苷酸序列测定

利用PCR技术 ,从含有大肠杆菌菌毛抗原K99基因的质粒pX5K中扩增出 0 .4kb的K99菌毛抗原基因 ,然后将其亚克隆到pGEM T easy载体上 ,并转化至受体菌JM10 9中 ,采用碱性裂解法提取质粒DNA后 ,经NcoI和EcoRⅠ双酶切分析和核苷酸序列分析 ,证明插入的K99基因片段具有正确的核苷酸序列     

含CpG motifs的乙肝表面抗原载体的构建及表达

构建了编码乙肝表面抗原中蛋白的卡那霉素抗性真核表达质粒pVAX-CpGS2S，其中通过PCR克隆了若干CpG motifs到质粒载体中。经酶切鉴定和测序正确后将重组质粒体外转染COS7细胞，ELISA法检测上清液中的HBsAg呈阳性，说明重组质粒pVAX-CpGS2S在体外能够有效表达HBsAg，为探讨CpG motifs克隆入DNA疫苗载体后对体内免疫反应的影响打下基础。     

壳聚糖修饰L-天门冬酰胺酶对酶活及抗原的影响

采用水溶性壳聚糖对L－天门冬酶胺酶进行化学修饰，修饰后的酶活回收率高达705，修饰酶的比活为每毫克蛋白质121.79U，pH值为7.4，更接近于人体血浆pH值(7.2-7.4），对底物的亲和力有所提高，稳定性增加，抗原性仅为自然酶的1／3，增加了临床使用的安全性。     

核仁抗原在白血病诊断中的初步观察

用抗核仁抗原NAg-1的抗血清对各种白血病人骨髓组织进行免疫荧光染色.结果表明,急性白血病患者的NAg-1阳性细胞率显著高于正常人及各种非白血病贫血患者.在处于完全缓解的白血病患者中,具有较高的NAg-1阳性率者(>10％)比较低者(<10％)更易复发.     

纳米铝佐剂诱导鸡提前产生抗AIVH9体液免疫应答

接种灭活油佐剂疫苗是目前国内防控禽流感主要的措施,为有效防控禽流感的流行发挥了重要的作用,但灭活苗首次接种后需要较长时间抗体才能达到有效保护水平.纳米颗粒具有独特的生物学特性,作为疫苗佐剂能引起机体强烈的免疫应答.本研究用纳米铝颗粒作为AIV H9的佐剂,结果显示,小鸡产生有效免疫保护抗体时间较常规油佐剂疫苗提前4天且无副反应,但抗体峰值和持续时间均低于油佐剂疫苗.氢氧化铝纳米颗粒作为佐剂可能对禽流感等重大传染病的紧急预防接种具有潜在的应用价值.
