对篮球运动中阵地进攻方式的运用研究

篮球运动阵地进攻在篮球运动中占有十分重要的位置，如何确立阵地进攻的基本体系、框架、以及整体运作的多种方式，就显得十分重要．本文力求从阵地进攻系统、方式、组合要素及结构优化等方面加以探讨，为确立篮球运动中阵地进攻的体系提出自己的观点。     

花特异表达启动子的克隆及花色调节基因Lc表达载体的构建

克隆了矮牵牛花特异表达基因CHSA启动子，并定向插入到已含有花色调节基因Lc的质粒pBI121中，取代原有的CaMV 35S启动子．所构建的新表达载体可用于花色改良研究．     

盐生杜氏藻cbr基因的克隆

首先对其他物种cbr／elip基因的同源序列进行相似性分析,设计一对简并引物,利用TR—PCR技术获得-250bp的片段,经克隆测序分析发现其同D．bardwail的cbr基因有67．0％的同源性,然后再以此片段为模板设计引物,通过RACE技术构建盐生杜氏藻cbr基因的全长序列．     

Gene cloning and molecular breeding to improve fiber qualities in cotton

Cotton fiber is one of known natural resources comprising the highest purity cellulose. It plays an important role world wide in the textile industry. With the acceleration of spinning speeds and the improvement of the people‘s living level, the demand of improving cotton fiber qualities is getting stronger and stronger. So, making clear the developmental model of fiber cell and elucidating systematically the molecular mechanisms of cotton fiber development and regulation will produce a great significance to make full use of cotton gene resources, raise cotton yield and improve fiber quality, and even develop man-made fiber. In the paper, the status of the gene cloning and the molecular breeding to improve cotton fiber quality were reviewed, the importance and potential of gene cloning related with cotton fiber quality were put forward and the proposal and prospect on fiber quality improvement were made. Using national resources available and through the creative exploration in corresponding research, some international leading patents in genes or markers linked with cotton fiber development having Chinese own intellectual property should be licensed quickly. And they can be used to improve cotton fiber quality in cotton breeding practice.     

Cloning of theAspergillus awamori glucoamylase gene and expression inSaccharomyces cerevisiae

The A. awamori glucoamylase I gene was obtained by RT_PCR and inserted into the plasmid pAC1. We constructed an expression vector pAC1_GA, which was used to transform the yeast S. cerevisiae AS 2 1364 without auxotrophic marker. The transformant secreted glucoamylase into the medium efficiently and degraded starch. The glucoamylase activity of the culture filtrate is up to 8 4 U/mL.     

盐藻3—磷酸甘油脱氢酶基因克隆

采用Ｌａｍｂｄａ置换型载体ＥＭＢＬ３构建了盐藻基因组文库，文库大小为１．８×１０４，插入片段大小为１０～２０ｋｂ，从盐藻基因组文库中获得３个３－磷酸甘油脱氢酶基因的阳性克隆，斑点杂交证实了原位杂交的真实性     

棉铃虫细胞系SFE—HA—8212的克隆化及对HaSNPV受纳性提高的克隆系的建立

用细胞临界生长密度法对棉铃虫蛹卵巢细胞系ＳＦＥ—ＨＡ—８２１２进行克隆化，获得８株克隆细胞系．各株克隆细胞系各自的形态较为均一，但相互间在形态、生长特性、对病毒受纳性等方面有很大差别，其中一株ＣＳ细胞对棉铃虫单粒包埋型核型多角体病毒（ＨａＳＮＰＶ）的受纳性明显高于ＳＦＥ—ＨＡ—８２１２细胞，形成多角体的细胞比率、多角体的产量、游离病毒粒子的产量及多角体蛋白的产量均有提高，是研究和应用ＨａＳＮＰＶ的有效系统．     

扬子鳄DMRT1基因DM保守区的克隆及序列分析

DMRT1基因是DMRT基因家族的一个重要成员,在动物性别决定过程中发挥重要作用.本文参照人DMRT1基因DM保守区及有关文献,设计了一对简并引物,扩增了扬子鳄的DMRT1基因,并对扩增产物进行了亚克隆与测序.结果在雌雄扬子鳄个体中各获得一个预期的基因片段,长度为140bp,序列无性别差异性,命名为扬子鳄AsDMRT1基因.序列同源性分析表明,扬子鳄DMRT1基因DM盒区核苷酸序列与人和鼠相应基因的同源性分别为87%、86%;编码的氨基酸序列与人和鼠相应基因编码序列的同源性分别为95%、97%.充分显示了DMRT1基因在进化上的高度保守性.     

Isolation and Characterization of Phytoene Desaturase cDNA from Stigma of Crocus sativus

Phytoene desatumse (PDS) has recently been identified as an important enzyme in carotenoid biosynthesis pathway. A cDNA clone encoding phytoene desaturase gene is isolated from stigma of saffron (Crocus sativus L. ) using RT-PCR technique. Sequence analysis shows 85% similarity to Narcissus pseudonarcissus, 79% to Zea mays, 78% to Arabidopsis thaliana, 77% to Lycopersicon esculentum. A new full-length cDNA is obtained by 5 ‘-RACE and 3‘ -RACE techniques. The cDNA is 2149bp long with an open reading frame of 1697bp, which encodes a polypeptide of 565 amino acids. Southern analy-sis shows that the PDS gene is a single copy in saffron. Northern blot analysis shows higher expression level of PDS gene in stigma and anther than in leaves and stem.     

红鲫促性腺激素α亚基的cDNA克隆

从红鲫(Red crucian earp)脑垂体中提取总RNA，根据鱼类促性腺激素(Gonadotropin Hormone，简称GTH)α亚基的保守性，设计并合成了一对20个碱基长的简并引物，用RT-PCR方法扩增并克隆了红鲫GTH-α亚基的两种不同的cDNA，分别命名为GTH-α1和GTH-α2。GTH-α1和GTH-α2分别由363和366个核苷酸组成，分别编码117和118个氨基酸．这两种类型的cDNA的核苷酸和氨基酸的同源性非常高，分别达到了95.0％和96．6％．但是，它们之间也有4个氨基酸的差别，而且GTH-α1有一个三联体核苷酸组成的氨基酸的缺失。聚类分析表明，GTH-α亚基在鲤科鱼类中具有高度保守性。
