用双极性膜电渗析法分离混合氨基酸   总被引:6,自引:0,他引:6  
为开发新的混合氨基酸分离技术,提出用双极性膜电渗析分离混合氨基酸的方法.在对混合氨基酸溶液中离子成分进行分析的基础上,预测了使用该法的可行性,并通过三室双极性膜电渗析法实验验证了理论计算的结果.对于等电点相差很大的谷氨酸和精氨酸以及等电点相差比较大的谷氨酸和甘氨酸以及精氨酸和甘氨酸两元混合物,可以实现很彻底的分离,得到纯度96%以上的产品,电流密度可以达到5~10 mA·cm-2. 对于等电点相差很小的丝氨酸和脯氨酸,则分离难度较大,电流密度在1 mA·cm-2以下.该法不需使用缓冲溶液,可提高分离过程的效率.  相似文献

Breast cancer still remains a major problem in its incidence, morbidity and mortality; therefore, more effective strategies for its prevention are urgently needed. Retinoids, natural and synthetic derivatives of vitamin A, possess antiproliferative and proapoptotic properties, making them a promising class of chemopreventive agents against breast cancer. The efficacy of all-trans retinoic acid, 9-cis-retinoic acid, LGD1069 (Targretin, bexarotene), and N-(4-hydroxyphenyl)retinamide (fenretinide) as breast cancer chemopreventive agents is being studied. A better understanding of the molecular mechanisms of action of these agents should lead to improvements in their clinical application. The immunosuppressants tacrolimus (FK506) and cyclosporin A (CsA) have increased the survival rates in organ transplantation. Both drugs inhibit the protein phosphatase calcineurin (CaN) in activated T cells, exhibiting similar side-effects. Diabetes is observed more often in FK506 than CsA therapy, probably due to inhibition of new molecular targets other than CaN. We studied FK506 toxicity in mammalian cells. FK506, but not CsA, regulated p38 activation by osmotic stress, and decreased viability in osmostressed cells. In addition, FK506 treatment strongly increased the phosphorylation of the eukaryotic initiation factor-2a (eIF-2a) subunit. eIF-2a phosphorylation, p38 inhibition and cell lethality were relieved by addition of excess amino acids to the medium, suggesting that amino acid availability mediated FK506 toxicity. Therefore, these FK506-dependent responses could be relevant to the non-therapeutic effects of FK506 therapy.Received 16 October 2003; received after revision 8 January 2004; accepted 14 January 2004  相似文献

Self-assembled structures of 3,4,5-tris-dodecy-loxy benzoic acid methyl ester (E12), 3,4,5-tris-tetradecy-loxy-benzoic acid methyl ester (E14) and their mixture (E12/E14) have been studied on HOPG by scanning tunneling microscopy (STM). Dimer-like patterns induced by dipole-dipole interaction are observed in E12 and E14 monolayers. The molecules form close-packed rows and interdigitate with the alkyl chains in adjacent molecules. The structural differences are proposed to be from the different length of substituted alkyl chains. Owing to similar adsorption energy, phase separation is observed in the E12 and E14 mixed adlayer with different domains.  相似文献

The carnitine system plays a key role in β-oxidation of long-chain fatty acids by permitting their transport into the mitochondrial matrix. The effects of hypothyroidism and hyperthyroidism were studied on γ-butyrobetaine hydroxylase (BBH), the enzyme responsible for carnitine biosynthesis in the rat. In rat liver, BBH activity was decreased in the hypothyroid state and increased in hyperthyroid animals. The modifications in BBH activity correlated with changes in the enzyme Vmax values. These changes were shown to be related to hepatic BBH mRNA abundance. Thyroid hormones are known to interact with lipid metabolism, in particular by increasing long-chain fatty acid oxidation through activation of carnitine-dependent fatty acid import into mitochondria. The effects and the relationship between sali-cylic acid(SA)and nitric oxide(NO) on Vicia faba L.stomatal movement were studied.The results here showed that exogenous SA and NO induced stomatal closure,100μmol/L SA induced a rapid and striking NO increase in the cytosol of guard cells.This phenomenon was largely prevented by 2000μmol/L 2-phenyl-4,4,5,5-tetramethylimidazoline-l-oxyl-3-oxide(PTIO),a specific NO scavenger,and 25μmol/L N^G-nitro-L-Arg-methyl eater (L-NAME),an inhibitor of NO synthase(NOS) in mammalian cells that also inhibits plant NOS.In addition,SA-induced stomatal closure was largely prevented by PTIO and L-NAME.These results provide evidence that guard cells generate NO in response to SA via NOS-like activity,and that such NO production is required for full stomatal closure in response to SA.H-(1,2,4)-oxadiazole-[4,3-α]quinoxalin-l-one(ODQ),an inhibitor of guanylate cyclase,and nicotinamide,an antagonist of cADPR production,inhibited the effects of SA-and NO-induced stomatal closure.It suggests that both cGMP and cADPR might mediate the signal transduction of SA and NO-induced stomatal closure.  相似文献

Low concentration of dithionite results in the reduction of Cu-Cn binuclear and heine a active sites of the cytochrome c oxidase thin solid film immersed in the acidic phosphate buffer, but Fe-Cu binuclear center keeps in the oxidation state. It manifests as a negative peak at 426 nm and a positive one at -408 nln in the difference spectra induced by dithionite. The former implies decrease of the oxidized form of heme a center, that is, Fea^3 →Fea^2 . And the latter results from the contribution of metal-ligand charge transfer (MLCT) transition in the reduced binuclear Cu-Cu cluster, rather than from that of heine a center. This stronger Soret MLCT band must be helpful to overcoming the difficulty in distinguishing the weaker copper sign from the stronger one of iron when studying copper-iron protein.  相似文献
